[cze][eng] V typické eukaryotické buňce tvoří mediátorová RNA (mRNA) asi 5 % celkové RNA, kde slouží jako předloha pro syntézu bílkovin. Nedávné výzkumy ukázaly, že mRNA jsou často transportovány do různých subcelulárních kompartmentů nebo cytoplazmatických oblastí, kde mohou být následně translatovány. Prostřednictvím lokalizace mRNA buňky regulují mnoho procesů, například vytváří lokální koncentraci specifických proteinů v reakci na aktuální nároky nebo určují interakčního partnera kódovaného proteinu. Právě z těchto důvodů lze považovat časoprostorovou distribuci mRNA za mezistupeň genové exprese. Tato práce se zaměřuje především na mechanismy transportu, které zprostředkovávají lokalizaci mRNA do cytozolických oblastí nebo membránových organel v méně diferencovaných a nepolarizovaných savčích buňkách. Recentní pokusy odhalily způsoby, jakými se transkripty shromažďují v protruzivních oblastech, prokázaly existenci několika druhů mRNA ohnisek a osvětlily význam lokalizace v centrozomech. Díky vysokoprůchodným zobrazovacím technologiím dnes známe lokalizační vzorce určitých typů transkriptů, avšak jestli se lokalizace týká i ostatních mRNA by mělo být předmětem dalších výzkumů. Recent researches have shown that lots of mRNAs are frequently transported to distinct subcellular compartments or cytoplasmic destinations, where they can subsequently undergo a localized translation. Through the localization cells regulate many processes, for instance they can generate a local concentration of specific proteins in response to the actual needs or determine an interacting partner of the encoded protein. Thus, spatiotemporal mRNAdistribution can be considered as an intermediate stage of gene expression. This thesis primarily focuses on transport mechanisms, that direct mRNA localization to cytosolic regions or membranelles organelles in less differentiated and non-polarized mammalian cells. Latest approaches discovered the ways in which transcripts accumulate in protrusions, demonstrated the existence of several mRNA foci and elucidated the importance of centrosome localization. To date high-throughput imaging techniques have revealed some remarkable localization patterns, but relatively a few transcripts has been studied yet, so if this could be applied to other mRNAs needs a further investigation.
