[cze][eng] Diplomová práce se zaměřuje na genetickou etiologii vrozených anatomických anomálií, včetně situs inversus a dalších přidružených patologií spojených s poruchami ciliárních funkcí, mezi které patří izomerie, heterotaxie nebo syndrom primární ciliární dyskineze. Popisuje anatomické dislokace orgánů typické pro jednotlivé diagnózy a jejich klinické projevy. V teoretické části se práce věnuje etiologii a patofyziologii těchto poruch, včetně jejich genetických aspektů. Praktická část zahrnuje analýzu genetického materiálu pacientů s těmito diagnózami pomocí sekvenování nové generace (NGS). Výsledky genetického vyšetření jsou dále diskutovány ve vztahu k fenotypovým projevům. Součástí práce jsou také kazuistiky vybraných pacientů. Klíčová slova: situs inversus, heterotaxie, dextrokardie, ciliární dysfunkce, sekvenování nové generaceThe thesis focuses on the genetic etiology of congenital anatomical anomalies, including situs inversus and other pathologies associated with ciliary function disorders, including isomerism, heterotaxy and primary ciliary dyskinesia syndrome. It describes the anatomical organ dislocations typical of each diagnosis and their clinical manifestations. The theoretical part of the thesis deals with the etiology and pathophysiology of these disorders, including their genetic aspects. The practical part includes the analysis of genetic material of patients with these diagnoses using next generation sequencing (NGS). The results of genetic testing are further discussed in relation to phenotypic manifestations. Case reports of selected patients are also included.
