[cze][eng] Neurofibromatóza typu I je autozomálně dominantní choroba způsobená mutacemi v genu NF1, který kóduje protein neurofibromin, tumorsupresor a významný regulátor mnoha buněčných procesů, včetně proliferace, karyokineze, organizace cytoskeletu a diferenciace nervových buněk. Prominentní vlastností této choroby je její mimořádná variabilní expresivita a široká škála klinických projevů. Markantní rozdíly mezi projevy nemoci lze pozorovat nejen mezi rodinami, ale i v rámci jedné rodiny, a to dokonce i u případů monozygotních dvojčat. Mezi faktory přispívající k variabilní expresivitě patří modifikující geny, posttranslační modifikace, alternativní sestřih, ale i epigenetické mechanismy. Konkrétní příčiny fenotypové variability jsou předmětem výzkumu již nejméně 30 let, dodnes však nejsou jednoznačně objasněny. Klíčová slova: neurofibromatóza, variabilní expresivita, genové interakce, gen NF1, neurofibrominNeurofibromatosis type I is an autosomal dominant disorder caused by mutations in the NF1 gene, which encodes the protein neurofibromin, a tumor suppressor and a key regulator of various cellular processes, including proliferation, karyokinesis, cytoskeletal organization, and neural cell differentiation. A prominent feature of this disorder is its extraordinary variable expressivity and a wide spectrum of clinical features. Striking differences in disease manifestation can be observed not only between families but also within a single family, even in monozygotic twins. Factors contributing to this variable expressivity include modifying genes, post-translational modifications, alternative splicing, and epigenetic mechanisms. The specific causes of phenotypic variability have been a subject of research for at least 30 years, but they remain incompletely understood to this day.
