Martin Žofka Název diplomové práce: Automatizace testu líhnutí vajíček vlasovky slezové Haemonchus contortus je známý parazit malých přežvýkavců, přičemž jeho přítomnost může vést k vážnému zhoršení zdravotního stavu zvířat a v krajních případech i k jeho úhynu. Boj proti H. contortus je komplikován vzrůstající anthelmintickou rezistencí, a proto je potřeba výzkumu a vývoje spolehlivých diagnostických metod k testování účinnosti anthelmintik. Tato diplomová práce se zaměřuje na porovnání citlivosti tří kmenů H. contortus (ISE, IRE a WR) k thiabendazolu (TBZ) pomocí testu líhnutí vajíček (Egg Hatch Test - EHT) a na porovnání manuálního a automatizovaného vyhodnocení testu. Dále byla testována účinnost albendazolu (ABZ) a jeho derivátu propargylalbendazolu (pABZ) na citlivém kmenu ISE. Výsledky naznačily, že automatizovaný systém může sloužit jako užitečný nástroj pro urychlení analýzy, avšak jeho přesnost není zatím tak vysoká jako u manuálního hodnocení.Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Martin Žofka Title of diploma thesis: Automation of Egg Hatch Test for Haemonchus contortus eggs Haemonchus contortus is a well-known parasite of small ruminants, and its presence can lead to a severe deterioration in the animals' health and, in extreme cases, even death. The fight against H. contortus is complicated by increasing anthelmintic resistance, highlighting the need for research and development of reliable diagnostic methods for testing anthelmintic efficacy. This thesis focuses on comparing the sensitivity of three H. contortus strains (ISE, IRE, and WR) to thiabendazole (TBZ) using the Egg Hatch Test (EHT), as well as comparing manual and automated test evaluation methods. In addition, the efficacy of albendazole (ABZ) and its derivative propargylalbendazole (pABZ) was tested on the susceptible ISE strain. The results suggested that the automated system may serve as a useful tool for speeding up the analysis; however, its accuracy is not yet as high as that of manual evaluation.
