Protein p53 je důležitým tumor-supresorovým transkripčním faktorem v eukaryotických buňkách. Prostřednictvím své aktivity má schopnost pozastavit buněčný cyklus v reakci na poškození genetické informace a získává pro buňku čas na opravu tohoto poškození. Mutace proteinu p53 se velmi často vyskytují v nádorových buňkách bez ohledu na typ postižené tkáně. Hlavním cílem zde předkládané diplomové práce bylo rekombinantní expresí připravit přirozenou formu proteinu p53 a jeho mutantní formy. Jedná se o mutantní formy p53, o kterých je známo, že ovlivňují jeho oligomerní stav, konkrétně se jedná o formy L344A a L344P. Dále bylo součástí práce charakterizovat interakci všech připravených proteinů s hemem. Pro expresi zmíněných proteinů byly použity buňky E. coli BL21 (DE3) transformované plasmidem kódujícím studovaný protein (pET21c-His-SUMO-p53WT, pET21c-His-SUMO-p53L344A nebo pET21c-His-SUMO-p53L344P). Pro izolaci a purifikaci všech tří proteinů byla použita niklová afinitní chromatografie, heparinová afinitní chromatografie a gelová permeační chromatografie. Úspěšnost těchto izolačních kroků byla posouzena elektroforézou na polyakrylamidovém gelu v prostředí dodecylsíranu sodného. The p53 protein is an important tumor-suppressor transcription factor in eukaryotic cells. Through its activity, it has the ability to arrest the cell cycle in response to genetic damage, thus providing the cell time to repair this damage. Mutations in the p53 protein are very frequently found in tumor cells regardless of the type of affected tissue. The main objective of the diploma thesis presented here was to prepare the wild type p53 protein and its mutant forms through recombinant expression. These mutant forms of p53 are known to affect its oligomeric state, specifically the L344A and L344P forms. Another part of this thesis involved characterizing the interaction of all prepared proteins with heme. For the expression of the mentioned proteins, E. coli BL21 (DE3) cells were transformed with a plasmid encoding the studied protein (pET21c- His-SUMO-p53WT, pET21c-His-SUMO-p53L344A, or pET21c-His-SUMO-p53L344P). Nickel affinity chromatography, heparin affinity chromatography, and gel permeation chromatography were used for the isolation and purification of all three proteins. The success of these isolation steps was assessed by polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate.
