Cílem této bakalářské práce je stanovení antimikrobiálních účinků plazmatem ošetřených kapalin na zvolené mikroorganismy. Plazmatem ošetřené kapaliny představují slibnou alternativu v boji proti bakteriálním infekcím, zejména v souvislosti s narůstající antibiotickou rezistencí. Přestože vykazují výrazné antimikrobiální účinky, jejich přesný mechanismus účinku dosud není plně objasněn, a proto tvoří hlavní předmět zkoumání této práce. Teoretická část je zaměřena na problematiku plazmatem ošetřených kapalin (PTL), včetně principu jejich produkce a přehledu systémů využívaných k jejich přípravě. Dále jsou zde popsány reaktivní částice v PTL, které mají největší podíl na antimikrobiální účinky. Součástí kapitoly je rovněž mikrobiologický přehled, zahrnující popis mikroorganismů vybraných pro experimentální část práce. Poslední část teoretické části se zabývá enzymy přítomnými v bakteriích a kvasinkách, které slouží jako ochrana proti účinkům reaktivních částic. V experimentální části byly plazmatem ošetřené kapaliny připraveny z fyziologického roztoku pomocí dvou plazmových systémů. Současně byly připraveny kontrolní roztoky simulující chemické složení a pH výsledných PTL. Testování účinků probíhalo na standardizovaných kulturách mikroorganismů. Mikrobiální suspenze byly vystaveny plazmatem ošetřeným kapalinám, roztokům simulujících složení a pH zkoumaných PTL. Ve zvolených expozičních časech bylo provedeno stanovení počtu životaschopných kolonií očkováním na živnou půdu a po 24hodinové inkubaci byl vyhodnocen antimikrobiální účinek. Výsledky této práce prokázaly vyšší antimikrobiální účinnost plazmatem ošetřených kapalin ve srovnání s roztoky simulujícími jejich složení a hodnotu pH. Zaznamenán byl rovněž rozdíl v účincích mezi dvěma různě připravenými plazmatem ošetřenými kapalinami. Dále bylo zjištěno, že Pseudomonas aeruginosa vykazuje nejvyšší citlivost k expozici PTL oproti ostatním zvoleným mikroorganismům.
The subject of this bachelor thesis is to evaluate the effects of plasma-treated liquids on selected microorganisms in order to assess their antimicrobial potential. Plasma-treated liquids represent a promising alternative in the fight against bacterial infections, particularly in the context of increasing antibiotic resistance. Although PTLs exhibit notable antimicrobial properties, the exact mechanisms of microbial inhibition have not been fully explained. Therefore they are the main focus of this study. The theoretical part of the thesis focuses on the issue of plasma-treated liquids (PTL), including the principles of their generation and an overview of the systems used for their production. Furthermore, the reactive species present in PTL are described, since these are primarily responsible for the antimicrobial efficacy. This section also includes an overview of microorganisms selected for the experimental part of the study. The final chapter of the theoretical section addresses the enzymes present in bacteria and yeasts that serve as protective mechanisms against the effects of reactive species. In the experimental part, plasma-treated liquids were prepared from physiological saline using two different plasma systems. Simultaneously, control solutions simulating the chemical composition and pH of the studied PTLs were prepared. The antimicrobial effects were tested on reference strains. Bacterial suspensions were exposed to the plasma-treated liquids, as well as to the studied control solutions. At selected exposure times, the number of culturable colonies was determined by standard plate count method. The results of this study demonstrated a higher antimicrobial efficacy of the plasma-treated liquids compared to the solutions mimicking their composition and pH. A difference in efficacy between the two PTLs prepared using different plasma systems was also observed. Furthermore, Pseudomonas aeruginosa was found to be the most sensitive to PTL compared to other the studied microorganisms.
