Cílem této bakalářské práce bylo optimalizovat metodu pro analýzu sterolů a terpenů v kávě pomocí vysokoúčinné kapalinové chromatografie (HPLC). Postupnou optimalizací byla jako stacionární fáze vybrána kolona Zorbax SB-C18 a jako mobilní fáze směs acetonitrilu a vody měnící poměry obou složek v čase. Pro detekci byl vybrán detektor s diodovým polem (DAD), sledovány byly vlnové délky 195, 220 a 290 nm, které byly identifikovány jako optimální pro jednotlivé analyty. Detekce pomocí hmotnostního detektoru nebyla účinná vzhledem k nepolárnímu charakteru studovaných látek a použití ionizace pomocí elektrospreje. Zatímco kafestol a kahweol se za použitých chromatografických podmínek podařilo účinně separovat, u studovaných sterolů díky jejich strukturní podobnosti tohoto cíle dosaženo nebylo. Optimalizovaná metoda byla použita pro analýzu kahweolu a kafestolu v reálných vzorcích kávy připravených různými baristickými metodami. Koncentrace kafestolu v kávě se v závislosti na použité baristické metodě pohybovala mezi 0,20 mg/l do 3,07 mg/l a koncentrace kahweolu 0,70 mg/l–11,5 mg/l.
The aim of this bachelor’s thesis was to optimize a method for the analysis of sterols and terpenes in coffee using high-performance liquid chromatography (HPLC). Through stepwise optimization, a Zorbax SB-C18 column was selected as the stationary phase, and a mixture of acetonitrile and water with time-dependent composition was chosen as the mobile phase. A diode array detector (DAD) was used for detection, monitoring wavelengths of 195, 220, and 290 nm, which were identified as optimal for the individual analytes. Detection using a mass spectrometric detector proved ineffective due to the non-polar nature of the studied compounds and the use of electrospray ionization (ESI). While cafestol and kahweol were successfully separated under the optimized chromatographic conditions, this was not achieved for the sterols due to their structural similarity. The optimized method was subsequently applied to the analysis of kahweol and cafestol in real coffee samples prepared using various barista techniques. The concentration of cafestol in the analyzed coffee samples ranged from 0.20 mg/l to 3.07 mg/l, while the concentration of kahweol ranged from 0.70 mg/l to 11.5 mg/l, depending on the preparation method used.
