Tato bakalářská práce byla zaměřena na charakterizaci termofilního mikroorganismu Caldimonas thermodepolymerans a jeho potenciál v oblasti biotechnologie nové generace, zejména při produkci a recyklaci polyhydroxyalkanoátů (PHA). Zkoumána byla schopnost produkce PHA z monomerních jednotek rozložitelných biopolymerů, jako jsou polyhydroxybutyrát, kyselina polymléčná, polybutylensukcinát a polykaprolakton. Dále byl sledován vliv vedlejšího produktu degradace PHA, kyseliny krotonové, na růst mikroorganismu. Výsledky ukázaly, že C. thermodepolymerans je schopna růstu a produkce PHA na 3-hydroxybutyrátu, přičemž kombinace 3-hydroxybutyrátu a xylózy vedla k vyššímu nárůstu biomasy a akumulace PHA. Při vyšších koncentracích kyseliny krotonové byl pozorován inhibiční účinek, avšak nižší koncentrace neomezovaly růst mikroorganismu. Sukcinát, kaprolakton a kyselina mléčná se v nastavených podmínkách ukázaly jako nevhodné substráty pro růst a produkci PHA tímto mikroorganismem. Získané výsledky potvrzují, že C. thermodepolymerans má potenciál pro aplikace v cirkulární biotechnologii.
This bachelor's thesis is focused on the characterization of the thermophilic microorganism Caldimonas thermodepolymerans and its potential in next-generation biotechnology, particularly in the production and recycling of polyhydroxyalkanoates (PHA). The study investigated the ability of the microorganism to produce PHA from monomeric units of biodegradable polymers such as polyhydroxybutyrate, polylactic acid, polybutylene succinate, and polycaprolactone. Additionally, the influence of crotonic acid, a by-product of PHA degradation, on microbial growth was examined. The results demonstrated that C. thermodepolymerans is capable of growth and PHA production on 3-hydroxybutyrate, with a combination of 3-hydroxybutyrate and xylose leading to increased biomass and PHA accumulation. Higher concentrations of crotonic acid showed an inhibitory effect, whereas lower concentrations did not limit microbial growth. Succinate, caprolactone, and lactic acid proved to be unsuitable substrates for growth and PHA production under the tested conditions. The findings confirm that C. thermodepolymerans holds promise for applications in circular biotechnology.
