Cieľom tejto bakalárskej práce bolo posúdiť kvalitu pôdy pomocou bioindikačných testov s využitím dážďovky hnojnej (Eisenia fetida) ako testovacieho organizmu. Skúmali sa účinky čistiarenského kalu a jeho kombinácie s biouhlím v artificiálnej pôde a pôde z reálneho prostredia. Testy hodnotili akútnu toxicitu a únikové správanie. Ďalšie testy, konkrétne subchronická expozícia a analýza biomarkerov oxidačného stresu – aktivita katalázy a koncentrácia TBARS (látky reagujúce s kyselinou tiobarbiturovou na stanovenie lipidovej peroxidácie), už boli hodnotené iba v rámci účinkov prídavku kalu do dvoch typov pôdy. Vyššie koncentrácie kalu viedli k vysokej mortalite a vyhýbaniu sa kontaminovanému prostrediu. Subchronický test odhalil pokles hmotnosti a zvýšenie obsahu proteínov v kontaminovaných pôdach, čo je v súlade so stresovou odpoveďou organizmu. Aktivita katalázy aj hodnoty TBARS vykazovali rozdielne výsledky v dvoch druhoch pôdy. Výsledky potvrdzujú význam komplexného hodnotenia toxických vplyvov s využitím viacerých úrovní biologickej odpovede.
The aim of this bachelor’s thesis was to assess soil quality using bioindication tests with the earthworm Eisenia fetida as the test organism. The effects of sewage sludge and its combination with biochar were examined in both artificial soil and soil from a real environment. The tests evaluated acute toxicity and avoidance behavior. Additional analyses, specifically subchronic exposure and biomarkers of oxidative stress — catalase activity and TBARS concentration (thiobarbituric acid reactive substances as an indicator of lipid peroxidation) — were assessed only for the effects of sludge addition in both soil types. Higher sludge concentrations led to high mortality and avoidance of the contaminated substrate. The subchronic test revealed a decrease in body weight and an increase in protein content in contaminated soils, consistent with a stress response. Catalase activity and TBARS levels showed differing results between the two soil types. The results highlight the importance of comprehensive assessment of toxic effects using multiple levels of biological response.
