Tato diplomová práce se zaměřuje na optimalizaci složení biokompozitu na bázi poly(3-hydroxybutyrátu) a kyseliny polymléčné, doplněného o keramické plnivo (hydroxyapatit a fosforečnan vápenatý) a změkčovadlo. Cílem práce bylo sledovat vliv různých poměrů těchto složek na výsledné mechanické vlastnosti jak porézní struktury scaffoldu, tak samotného materiálu a dále vyhodnotit jeho degradabilitu v podmínkách simulujících prostředí lidského těla. Pro určení mechanických vlastností byla provedena zkouška v tahu a tlaku a degradace vzorků byla pozorována především pomocí hmotnostního úbytku a změny molekulové hmotnosti. Výsledky naznačují, že během degradace docházelo ke štěpení polyesterových řetězců a největší vliv na degradaci měl poměr složek P3HB/PLA. Přínosem této práce je komplexní pohled na chování biokompozitních materiálů nejen z hlediska mechanických vlastností, ale i jejich dlouhodobé degradace, což může sloužit jako podklad pro návrh scaffoldových struktur využitelných v kostním tkáňovém inženýrství.
This thesis focuses on the optimization of a biocomposite formulation based on poly(3-hydroxybutyrate), polylactic acid, incorporated ceramic fillers (hydroxyapatite and calcium phosphate) and a plasticizer. The aim of this work was to investigate the effect of various ratios of these components on the resulting mechanical properties of both the porous scaffold structure and material itself. This work also evaluates the material’s degradability under conditions simulating the human body environment. To evaluate the mechanical properties, tensile and compressive tests were performed. Degradation was monitored mainly through mass loss and changes in molecular weight. The results suggest that the hydrolytic degradation proceeded through the cleavage of polyester chains, with the P3HB to PLA ratio being the most critical factor affecting the rate of degradation. The main contribution of this work is a comprehensive insight into the behaviour of biocomposite materials in terms of both mechanical performance and long-term degradation, providing a solid basis for the design of scaffold structures used in bone tissue engineering.
