Tato diplomová práce se zabývá přípravou alginátových hydrogelů a studiem vlivu různých podmínek přípravy na jejich vnitřní strukturu a morfologii. Hydrogely představují atraktivní skupinu materiálů díky své schopnosti zadržovat vodu, biokompatibilitě a variabilitě vlastností, které je možné navíc i cíleně řídit volbou kompozice daného materiálu a modifikací jeho vnitřní struktury. Cílem práce bylo připravit alginátové hydrogely za různých procesních podmínek, analyzovat jejich morfologické a mechanické charakteristiky a posoudit, jak tyto podmínky ovlivňují výsledné vlastnosti hydrogelu. Experimentální část byla zaměřena na studium vlivu teploty mražení a doby, po kterou je vzorek hydrogelu mražen před navazujícím lyofilizačním krokem. Výsledky byly vyhodnoceny pomocí rastrovací elektronové mikroskopie (SEM), BET analýzy, reologických měření a bobtnacích testů. Získané poznatky přispívají k optimalizaci přípravy hydrogelů pro jejich potenciální agronomické využití jako nosičů půdních rhizobakterií.
This diploma thesis focuses on the preparation of alginate hydrogels and the study of the influence of various preparation conditions on their internal structure and morphology. Hydrogels represent an attractive group of materials due to their water retention capacity, biocompatibility, and variability of properties. The aim of the thesis was to prepare alginate hydrogels under different process conditions, analyse their morphological and mechanical characteristics, and assess how these conditions affect the final properties of the hydrogels. The experimental part was focused on studying the influence of temperature of freezing and the duration of the freezing step, which is important before the subsequent freeze-drying step. The results were evaluated using scanning electron microscopy (SEM), BET analysis, rheological measurements, and swelling tests. The findings contribute to the optimization of hydrogel preparation for their potential agronomic application as carriers of soil rhizobacteria.
