Alzheimerova choroba je podle Světové zdravotnické organizace nejčastější formou demence, která vzniká v důsledku úbytku mozkových buněk. Zatímco dříve postihovala především lidi starší 65 let, dnes se stále častěji objevuje i u čtyřicátníků. První příznaky se mohou projevit po 10 až 20 letech po zasažení mozku Alzheimerovou chorobou. Mezi spouštěče této nemoci odborníci řadí dědičnost, vlivy životního prostředí, virové a bakteriální infekce. Jedním z klíčových znaků je hromadění amyloidu beta, který svým hromaděním poškozuje nervové buňky, což vede k jejich trvalému odumírání. V této práci jsme se věnovali přírodním látkám extrahovaných z běžně dostupných doplňků stravy, které mají podpořit kognitivní funkce. Cílem této práce bylo stanovit vliv ženšenu, korálovce ježatého a apigeninu na viabilitu buněčných linií MCF-7, ND7/23, C57 BL6 a U87-MG. To bylo provedeno MTT testy, kde byla určena hodnota LD50 každé vybrané přírodní látky na jednotlivé buněčné linie zvlášť. Obecně nejodolnějšími buňkami byly buňky U87-MG, které dosáhly hodnoty LD50 pouze v přítomnosti 25 M apigeninu. Z testovaných buněčných linií byly ND7/23 nejcitlivější, protože u nich došlo k poklesu viability pod 50 % jako u prvních při dané koncentraci přírodních látek. Z testů viability byly vybrány vhodné koncentrace pro další měření, a tedy další z cílů této diplomové práce, ověření vlivu vybraných látek na expresi proteinů spojených s výskytem Alzheimerovy choroby. Jako proteinové markery tohoto onemocnění byly vybrány TAU, IL-6, IL-1, TNF, Aktin- a INF. Sledování změn exprese vybraných markerů bylo provedeno pomocí RT-qPCR. Buněčné linie ND7/23 a U87-MG byly kultivovány s vybranými látkami a A po dobu tří a šesti dnů. Pouze v případě ženšenu došlo po šesti dnech kultivace k signifikantně významnému poklesu exprese proteinu Tau a Il-6 pro buněčnou linii ND7. V případě buněk U87-MG nebyly zjištěny signifikantně významné hodnoty, ale pro korálovce ježatého v šestý den kultivace bylo pro proteinu Tau naměřeno mírné zvýšení Ct hodnoty, tedy snížení exprese Tau proteinu. Snížení exprese těchto proteinu již po šesti dnech kultivace poukazují na možnost využití ženšenu a korálovce ježatého pro případnou prevenci Alzheimerovy choroby a další výzkum s dlouhodobou kultivací.
The World Health Organization states that Alzheimer's disease is the most common cause of dementia, which is triggered by the degeneration of brain cells. It primarily affects individuals aged 65 and older, but in recent years, it has increasingly been observed in population of people around the age of 40. The first symptoms can appear 10 to 20 years after diagnosis. Experts consider genetic predisposition, environmental factors, and viral or bacterial infections among the triggers of Alzheimer's disease. One of the key hallmarks is the accumulation of betaamyloid, which damages nerve cells through its buildup, eventually leading to their permanent death. 4 In this study, we focused on natural compounds extracted from commonly available dietary supplements that are believed to support cognitive functions. The aim of this work was to determine the effect of ginseng, lion’s mane mushroom (Hericium erinaceus), and apigenin on the viability of MCF-7, ND7/23, C57 BL6, and U87-MG cell lines. This was conducted using MTT assays, where the LD50 value of each selected natural substance was determined for each cell line separately. In general, the most resistant cells were U87-MG, which reached the LD50 value only in the presence of 25 M apigenin. The least resistant were the ND7/23 cells, which reached LD50 the earliest among all tested lines. Based on the viability tests, suitable concentrations were selected for further measurements and thus another objective of this thesis: to evaluate the impact of the selected compounds on the expression of proteins associated with Alzheimer's disease. The selected protein markers for this condition were TAU, IL-6, IL-1, TNF, Actin-, and IFN. Changes in expression of the selected markers were monitored using RT-PCR. The chosen substances were cultured with ND7/23 and U87-MG cell lines for three and six days in the presence of A. Only in the case of ginseng there was a statistically significant decrease in Tau and IL-6 protein expression observed after six days of cultivation in the ND7 cell line. In the U87-MG cells, no statistically significant results were found; however, for lion’s mane mushroom on the sixth day of cultivation, a slight increase in the Ct value for the Tau protein primer was measured, indicating a decrease in Tau protein expression. This opens further possibilities for future testing
