Táto diplomová práca sa zaoberá štúdiom synergického efektu antimikrobiálnych látok, ich enkapsuláciou do kombinovaných nanovlákien a zapracovaním do emulzných gélov s cieľom vyhodnotiť ich potenciál pre kozmetické a farmaceutické aplikácie. V teoretickej časti boli charakterizované nanomateriály, najmä nanovlákna, ďalej hydrogély, emulzné gély a kombinované nanomateriály. Následne boli popísané antimikrobiálne látky, mechanizmy ich účinku a antibiotická rezistencia. Pozornosť bola venovaná tiež synergickým testom a antioxidantom. Posledná časť obsahuje prehľad testov biokompatibility. V experimentálnej časti boli otestované štyri rastlinné antimikrobiálne látky (berberín, eugenol, karvakrol, tymol) a tri antibiotiká (erytromycín, gentamicín, streptomycín) z hľadiska ich antioxidačného efektu, minimálnej inhibičnej a letálnej koncentrácie. Na hodnotenie antimikrobiálneho účinku boli využité dve grampozitívne baktérie, a to Cutibacterium acnes a Staphylococcus epidermidis a gramnegatívna baktéria Escherichia coli. Následne boli vytvorené kombinácie látok za účelom stanovenia synergických interakcií, pričom celkovo išlo o 18 dvojíc látok. Najsilnejšie synergické účinky boli pozorované pri kombináciách berberín – tymol, karvakrol – eugenol a berberín – streptomycín. Tieto kombinácie boli následne enkapsulované do viacvrstvových nanovlákien, ktoré vykazovali vysokú mieru antioxidačnej aj antimikrobiálnej aktivity. Synergicky pôsobiace prírodné látky boli aplikované aj do emulzných gélov, ktoré však preukázali podstatne nižšiu účinnosť. Testy biokompatibility potvrdili, že pripravené kombinované nanovlákna sú netoxické a vhodné na kožné aplikácie. Výsledky práce poukazujú na potenciál využitia pripravených materiálov v oblasti kozmetiky a farmácie, napríklad vo forme pleťových masiek, náplastí na akné či materiálov na krytie rán. Zároveň synergické kombinácie môžu predstavovať účinný nástroj v prevencii vzniku rezistencie na antibiotiká.
This diploma thesis focuses on the study of the synergistic effect of antimicrobial agents, their encapsulation into composite nanofibers, and incorporation into emulsion gels with the aim of evaluating their potential for cosmetic and pharmaceutical applications. The theoretical section covered nanomaterials, specifically nanofibers, hydrogels, emulsion gels, and composite nanomaterials. It also discussed properties and mechanisms of action of antimicrobial agents, as well as the issue of antibiotic resistance. Further attention was given to synergy testing and antioxidants. The last section provided an overview of biocompatibility testing methods. In the experimental part, four plant-derived antimicrobial agents (berberine, eugenol, carvacrol, thymol) and three antibiotics (erythromycin, gentamicin, streptomycin) were tested for their antioxidant properties, minimum inhibitory concentration, and minimum lethal concentration. The antimicrobial activity was tested against two Gram-positive bacteria, namely Cutibacterium acnes and Staphylococcus epidermidis and Gram-negative bacteria Escherichia coli. Eighteen combinations were tested for synergistic interactions, with the strongest effects observed in the pairs berberine – thymol, carvacrol – eugenol, and berberine – streptomycin. These combinations were encapsulated into multilayer nanofibers, which demonstrated strong antioxidant and antimicrobial properties. When incorporated into emulsion gels, the synergistic compounds displayed significantly lower efficacy. Biocompatibility tests confirmed that the prepared composite nanofibers are non-toxic and suitable for skin applications. The findings highlight the potential of the developed materials for cosmetic and pharmaceutical applications, such as facial masks, acne patches, or wound dressings. Moreover, the synergistic combinations may represent an effective tool in preventing the development of antibiotic resistance.
