Předložená diplomová práce se zabývá řešením problematiky migrace sulfapyridinu v půdních systémech. Hlavním cílem této práce je navrhnout jednoduchý matematický model na základě experimentálních výsledků difúze léčiva v agarovém gelu v přítomnosti huminových látek jako hlavní složky organické hmoty v půdě. V dané magisterské práci byly provedené simulace transportu sulfapyridinu v pórovitém prostředí, ovlivněné vedlejšími chemický reakcí, imobilizujícími léčivo. Pro tyto výpočty bylo použité softwarové prostředí COMSOL Multiphysics a příslušné moduly „Chemistry“ a „Transport of diluted species in porous media“, umožňující simulace reálných procesů. Pomocí parametrické studie byly v této práce identifikované klíčové veličiny: difuzní koeficient na hodnotu D=4,7510-7 cm2/s, rychlostní konstanta přímé reakce (sorpce/imobilizace) byla stanovená na hodnotu Rkf=110-3 m3/(smol), a rychlostní konstanta zpětné reakce (nebo desorpci) na hodnotu Rkb=510-4 1/s. Model prokázal dobrou shodu s experimentálními data, což potvrzuje vhodnost nastavených parametrů. Navržený postup poskytuje robustní metodiku pro simulaci transportu farmaceutik v environmentálních matricích a napomáhá predikci šíření kontaminantů v půdě.
This thesis deals with the problem of sulfapyridine migration in soil systems. The main objective of this thesis is to propose a simple mathematical model based on experimental results of drug diffusion in agar gel in the presence of humic substances as the main component of organic matter in soil. In the present master thesis, simulations of sulfapyridine transport in porous media, influenced by chemical side reactions, and drug mobilization were carried out. For these calculations, the COMSOL Multiphysics software environment and the corresponding modules “Chemistry” and “Transport of diluted species in porous media” were used, allowing simulations of real processes. Using a parametric study, the key variables identified in this work were: the diffusion coefficient was set at D=4,7510-7 cm2/s, the rate constant of the direct reaction (sorption/immobilization) was set at Rkf=110-3 m3/(smol), and the rate constant of the reverse reaction (or desorption) was set at Rkb=510-4 1/s. The model showed good agreement with the experimental data, confirming the suitability of the set parameters. The proposed procedure provides a robust methodology for simulating the transport of pharmaceuticals in environmental matrices and aids in the prediction of contaminant dispersion in the soil.
