Tato diplomová práce se zabývá přípravou hybridních gelových sítí na bázi pol(yethylen glykolu) (PEG). Pro přípravu byl zvolen poly(ethylen glykol) diakrylát (PEGDA), který sloužil jako základ hydrogelové matrice, do které byly následně inkorporovány další komponenty za vzniku semi-interpenetrujících (semi-IPN) a interpenetrujících (IPN) hydrogelů. Pro inkorporaci do hydrogelové matrici na bázi PEG byly vybrány tři polyelektrolyty a to poly(4-styrensulfonát) sodný (PSS), diethylaminoethyl (DEAE) dextran a alginát sodný (SA). U semi-IPN hydrogelů byly připraveny celkem tři různé koncentrace (0,001; 0,01; 0,1 hm. %) pro každý polyelektrolyt. Pro IPN hydrogely byl aplikován pouze SA o koncentracích 0,5 a 1 hm. %, který byl síťován vápenatými ionty. Pro charakterizaci mechanických vlastností vzniklých hydrogelů byla v případě semi-IPN hydrogelů využita reologie, dále bylo analyzováno jejich botnání, transparentnost, transportní vlastnosti prostřednictvím jednoduchých difúzních experimentů. Průměrné hodnoty komplexních modulů, které byly získány za využití amplitudového oscilačního smykového testu, jednoznačně prokázaly, že u semi-IPN hydrogelů s inkorporovaným PSS a SA nedochází k ovlivnění viskoelasticity gelů přítomností semi-interpenetrovaných složek. V případě hydrogelu s DEAE-dextranem o koncentraci 0,1 hm. % došlo ke statisticky významnému poklesu hodnot komplexního modulu a tím ovlivnění mechanických vlastností. U botnání nedošlo k signifikantní změně přídavkem polyelektrolytu, stejně tak i v případě transparentnosti nebyly pozorovány významné rozdíly. Pro sledování transportních vlastností byly využity dvě metody, vizuální sledování prostupu modelové látky pomocí transiluminátoru, a dále kvantitativní metoda konstantního zdroje spojená se stanovením efektivního difuzního koeficientu modelové aktivní ingredience (nabitá aromatická organická barviva). Obě metody potvrdily ovlivnění transportních vlastností u hydrogelů s inkorporovaným PSS, a proto tento typ hydrogelu byl podroben i morfologické analýze pomocí rastrovací elektronové mikroskopie. Analýza potvrdila, že přídavek PSS do struktury hydrogelu nemá vliv ani na vnitřní architekturu gelu. Pro charakterizaci mechanických vlastností IPN hydrogelů na bázi PEG a SA byl využit amplitudový oscilační smykový test, který odhalil snížení hodnot komplexního modulu. Dále byly IPN hydrogely podrobeny botnací analýze, která prokázala sníženou schopnost botnat u nejvyššího obsahu SA v gelu (1 hm. %). V rámci sledování transparentnosti nedošlo k pozorování odlišností v porovnání s ostatními připravenými vzorky.
This master's thesis deals with preparation of hybrid gel networks based on poly(ethylen glycol) (PEG). For the preparation was chosen poly(ethylene glycol) diacrylate (PEGDA), which was used hydrogel to provide by radical crosslinking a chemical network hydrogel matrix, into which other components were incorporated to form semi-interpenetrating (semi-IPNs) and interpenetrating (IPNs) hydrogels. Three polyelectrolytes were selected for incorporation into the PEG-based hydrogels matrix, namely sodium poly(4-styrenesulfonate) (PSS), diethylaminoethyl (DEAE) dextran and sodium alginate (SA). For semi-IPNs hydrogels were prepared three different concentrations (0.001; 0.01 and 0.1 wt. %) for each of the polyelectrolytes. For IPNs hydrogels was applied only SA, which was crosslinked with calcium(II) ions, with concentrations of 0.5 and 1 wt. %. For characterisation of semi-IPNs hydrogels was used rheology to monitor mechanical properties, complemented by swelling analysis, transparency monitoring an determination of d transport properties via diffusion experiments. The average values of complex moduli, obtained using the amplitude oscillation shear test, clearly demonstarted the mechanical properties of semi-IPNs hydrogels were not affected by the presence of incorporated PSS and SA. In the case of hydrogels with DEAE-dextran at a concentration of 0.1 wt. %, there was a statistically significant decrease in the values of the complex modulus and affecting the mechanical properties. No significant changes in swelling was caused by the tested additions of polyelectrolytes. No significant differences were observed in the case of transparency monitoring either. Two methods were used to monitor the transport properties, visual monitoring of the model substance using a transilluminator and quantitative determination of the effective diffusion coefficient using the constant source diffusion method. Both approaches confirmed the influence of the incorporated PSS on the transport properties of hydrogels, and therefore this type of hydrogel was subjected to morphological analysis using scanning electron mocroscopy. The analysis confirmed that the addition of PSS to hydrogel structure does not affect the ultrastructure of the gel. For characterization of mechanical properties of IPNs hydrogel based on PEG and SA was used an amplitude oscillation shear test, which revealed a decrease in the values of the complex modulus. IPNs hydrogels were also subjected to swelling characteristics, which showed a reduced ability to swell caused by the highest content (1 wt. %) of SA. No differences in the transparency were observed compared to the other prepared samples.
