Cílem této diplomové práce bylo připravit nosičové systémy pro antibiotikum Ciprofloxacin, založené na hyaluronanu, jehož hydrofobní domény byly zpřístupněny za účelem efektivního navázání antibiotika (léčiva). Tento proces byl realizován pomocí ko-rozpouštědla (TBA) a vhodně zvoleného lyofilizačního programu, který umožnil optimalizaci přípravy nosičových systémů a zpřístupnění hydrofobních domén pro interakci s léčivem prostřednictvím hydrofobního efektu. Druhým hlavním směrem této práce byl pokus o maskování připravených nosičových systémů pomocí syntetických polymerů, konkrétně nanesením polyethylenglykolu (PEG), což mělo za cíl ochranu nosičového systému před eliminací imunitním systémem. Tento krok má potenciál znemožnit navázání sérového albuminu na nosičový systém, což by vedlo k jeho rychlé eliminaci z organismu. V rámci diplomové práce byla vypracována literární rešerše zaměřená na současné trendy v oblasti přípravy nosičových systémů léčiv, včetně přehledu instrumentálních technik, které byly použity pro analýzu připravených systémů. Experimentální část práce byla rozdělena na dvě hlavní fáze: přípravu nosičových systémů na bázi hyaluronanu a léčiva (Ciprofloxacinu) a podrobné studium připravených nosičových systémů. Morfologické charakteristiky připravených systémů byly studovány pomocí rastrovací elektronové mikroskopie (SEM), zatímco fyzikálně-chemické vlastnosti, včetně interakcí mezi hyaluronanovými řetězci a antibiotikem, byly zkoumány pomocí: infračervené spektrometrie s Fourierovou transformací, excitačně-emisních spekter a steady-state fluorescenční spektrometrie. Tato práce má potenciál přispět k lepšímu pochopení interakcí mezi přírodními polymery a farmaceutickými látkami a otevřít cestu k vývoji účinnějších nosičových systémů pro cílené podávání léčiv. The aim of this thesis was to prepare carrier systems for the antibiotic Ciprofloxacin, based on hyaluronan, whose hydrophobic domains were made accessible for the effective binding of the antibiotic (drug). This process was carried out using a co-solvent (TBA) and a suitably selected lyophilization program, which enabled the optimization of carrier system preparation and the exposure of hydrophobic domains for interaction with the drug through the hydrophobic effect. The second main focus of this work was an attempt to mask the prepared carrier systems using synthetic polymers, specifically by applying polyethylene glycol (PEG), aimed at protecting the carrier system from elimination by the immune system. This step has the potential to prevent serum albumin from binding to the carrier system, which would lead to its rapid elimination from the body. A literature review was developed as part of this thesis, focusing on current trends in the preparation of drug carrier systems, including an overview of the instrumental techniques that were used for the analysis of the prepared systems. The experimental part of the work was divided into two main phases: the preparation of hyaluronan-based carrier systems with the drug (Ciprofloxacin) and the detailed study of the prepared carrier systems. The morphological characteristics of the prepared systems were studied using scanning electron microscopy (SEM), while the physicochemical properties, including interactions between hyaluronan chains and the antibiotic, were examined using Fourier transform infrared spectroscopy (FTIR), excitation-emission spectra, and steady-state fluorescence spectroscopy. This work has the potential to contribute to a better understanding of the interactions between natural polymers and pharmaceutical compounds and open the way for the development of more efficient drug delivery systems for targeted drug delivery.
