Diplomová práce se zabývá analýzou senzoricky aktivních látek ječného sladu metodou HS-SPME-GC-MS. Byly analyzovány vzorky světlého sladu plzeňského typu, světlého karamelového sladu a barvicího čokoládového sladu, čtyř sladovnických odrůd ječmene – LG TOSCA, RGT PLANET, LG STAMGAST a BOJOS. Teoretická část práce je zaměřena na vznik senzoricky aktivních látek sladu. Jsou popsány obsahové látky ječmene, technologie sladování a chemické reakce vedoucí k tvorbě chuťových a vonných látek. V experimentální části práce byly optimalizovány podmínky metody extrakce HS-SPME. Za těchto optimalizovaných podmínek byly senzoricky aktivní látky extrahovány použitím vlákna 50/30 m DVB/CAR/PDMS a analyzovány metodou GC-MS. Separace látek probíhala na dvou chromatografických kolonách – Rxi-5ms a Supelcowax 10. Byly porovnány senzorické profily jednotlivých druhů sladů a popsány rozdíly mezi nimi. Práce zhodnotila vliv odrůdy, zeměpisného původu a technologie zpracování. Výsledky ukazují, že složení a zastoupení senzoricky aktivních látek světlého sladu ovlivňuje zejména odrůda a zeměpisný původ ječmene, zatímco u speciálních sladů převažuje vliv technologického procesu výroby.
The master's thesis deals with the analysis of sensory-active compounds in barley malt using the HS-SPME-GC-MS method. Samples of pale Pilsner-type malt, pale caramel malt and chocolate malt, produced from four malting barley varieties - LG TOSCA, RGT PLANET, LG STAMGAST and BOJOS were analysed. The theoretical part of the thesis is focused on the formation of sensory-active compounds in malt. Chemical components of barley, malting process and chemical reactions leading to the formation of flavour and aroma substances are described. In the experimental part, the conditions of the HS-SPME extraction method were optimised. Under these optimised conditions, sensory-active compounds were extracted using a 50/30 m DVB/CAR/PDMS fiber and analysed by GC-MS method. Two chromatographic columns, Rxi-5ms and Supelcowax 10, were used for the separation of analytes. The sensory profiles of the different malt types were compared and the differences between them were described. The influence of barley variety, geographical origin and malting technology was evaluated. The results indicate that in pale malt, the composition and abundance of sensory-active compounds is primarily influenced by the barley variety and its geographical origin, whereas in specialty malts, the influence of technological process of production is predominant.
