THESIS SYNOPSIS This thesis focuses on the characterization of RNA binding proteins (RBPs) that are intricate in the selection and translation of mRNA harbouring cis regulatory elements to guide the male and female gametophytes development. The eukaryotic translation initiation complex (eIF3) is contemplated to be the most prevalent to function in both sporophytic and gametophytic development. The major goal of this dissertation is to investigate the structural regulation and mechanisms of eIF3 complex subunit E (eIF3E) in controlling plant fertility. We show that eIF3E regulate mRNA translation via enhancer-repressor motifs combination to maintain normal pollen development. Further, this dissertation demonstrate that eIF3E-PCI domain plays a crucial role in eIF3E dissociation post translation initiation and simultaneously engages eIF3E assembly with the proteasome supercomplex for protein quality surveillance. We provide evidence that PCI domain situated phosphorylation sites, Thr417 and Ser421, are critical for eIF3E interaction with eIF3L accessory subunit and modulate the translational capacity of eIF3E-dependent cis-motifs containing mRNA translatability.
