CZ Neuroblastom (NBL), maligní embryonální nádor vycházející z periferního nervového systému, představuje nejčastější solidní extrakraniální nádor u dětí. Jeho závažnost a často nepříznivá prognóza podtrhují naléhavou potřebu vývoje nových a účinnějších léčebných strategií v onkologii. Navzdory pokroku v oblasti léčby nádorových onemocnění zůstává léková rezistence významným problémem. Jedním z mechanismů přispívajících k lékové rezistenci je lyzozomální sekvestrace. V tomto procesu se určitá léčiva, zejména slabé báze s hydrofobními vlastnostmi, akumulují v lyzozomech, buněčných organelách s kyselým prostředím udržovaným protonovou pumpou V-ATPázou. Tato sekvestrace brání lékům v dosažení jejich cílů v buňce, čímž je činí neúčinnými. Tato dizertační práce si klade za cíl provést analýzu exprese podjednotek V-ATPázy v buňkách NBL, odhalit případné abnormality a objasnit jejich roli v rozvoji rezistence vůči léčbě ellipticinem a cisplatinou. U buněk NBL rezistentních vůči ellipticinu a cisplatině byla pozorována zvýšená exprese podjednotky ATP6V0D1 V-ATPázy. NBL buňky také vykazovaly zvýšenou lyzozomální kapacitu. Jak inhibice funkce V-ATPázy, tak snížení exprese ATP6V0D1 (knockdown) zvýšily citlivost buněk na ellipticin a cisplatinu. Knockdown ATP6V0D1 dále potlačil proliferaci a migraci, snížil... EN Neuroblastoma (NBL) is a malignant embryonic tumor that originates from the peripheral nervous system. It is the most common solid extracranial tumor in children. The severity of neuroblastoma and the often unfavorable prognosis highlight the urgent need for the development of new and more effective cancer treatments. Notwithstanding the advancements that have been made in the field of oncology, the issue of drug resistance remains a significant challenge. One mechanism that contributes to the development of drug resistance is lysosomal sequestration. In this process, specific drugs, particularly weak bases with hydrophobic properties, accumulate in lysosomes, which are cellular organelles with an acidic environment maintained by the V-ATPase proton pump. This process of sequestration prevents the drugs from reaching their intended cellular targets, thereby rendering them ineffective. The objective of this dissertation is to conduct a comprehensive examination of V-ATPase subunit expression in NBL cells, identify potential abnormalities, and elucidate their role in the development of resistance to ellipticine and cisplatin treatment.
