[cze][eng] Pokročilé biofyzikální metody využívají fluorescenční značky navázané na studova-ný analyt a dokáží tak účinně podávat informace o tomto materiálu zájmu. Tato diplomová práce se věnuje fluorescenčnímu značení v biologických tkáních a skládá se ze dvou částí. První část, teoretická, shrnuje základní poznatky týkající se luminiscenčních jevů a jeho podskupin. Luminofory jsou popsány na základě 3 úrovní, na první úroveň je stavěna přirozená luminiscence fluorescenčních proteinů, následují organická barviva a poslední část charakterizuje nejmodernější techniky fluorescence pomocí nanomateriá-lů, jako jsou kvantové tečky (QDs). Experimentální část je zacílena na kvantifikaci kvantových teček, konkrétně byly použity karboxylované kvantové tečky (λem = 655 nm) v biokonjugaci s protilátkou proti proteinu p53. Tento biokonjugát je vysoce afinitní k proteinu p53 a díky tomu je možné dále stanovit množství proteinu p53 ve vzorku (například pomocí kapilární elektroforézy s laserem indukovanou fluorescenční detekcí či fluorescenční spektrome-trie) nebo pomocí fluorescenční mikroskopie monitorovat jeho distribuci v buňkách či tkáních.Advanced biophysical methods utilize fluorescent labels coupled to the analyte under study to report this material of interest effectively. This thesis is devoted to fluorescent labelling in biological tissues and consists of two parts. The first, theoretical, part summarizes the basic knowledge of the luminescence phenomena and its subgroups. Luminophores are described based on 3 levels: natural luminescence of fluorescent proteins being on the first level, organic dyes representing the second one and the last one mentioning the most up-to-date techniques of flu-orescence techniques using nanomaterials, like quantum dots (QDs). The experimental part focuses on the quantification of quantum dots. Specifically, carboxylated quantum dots (λem = 655 nm) were used in this part for the bioconjugati-on with an antibody against p53 protein. This bioconjugate has a high affinity for the p53 protein, and thus it is possible to determine the amount of p53 protein in the sample (e.g. by fluorescence spectrometry or capillary electrophoresis using laser-induced flu-orescence detection) or monitor its distribution in cells or tissues by fluorescence microscopy.
