[cze][eng] Procesem RNA interference je regulována transkripce a vnitrobuněčná exprese genů, kdy jsou specifické fragmenty dvoušroubovicové RNA zapojeny do indukce sekvenčně specifického potlačení genové exprese homologní mRNA. RNA interference je přítomna u všech eukaryotických organismů a je tedy považována za evolučně starý mechanismus. Metody RNA interference jsou dnes považovány za významné a zároveň rychlé technologie, kterými je možné provádět funkční analýzy genů a představují důležitou metodu jak v základním výzkumu, tak i v terapii mnohých nemocí. Práce byla zaměřena na testování účinnosti různých transfekčních reagencií a postupů vedoucích k optimálním podmínkám pro umlčení požadovaného genu metalothioneinu. Pro experimenty byly využity tři komerčně dostupné transfekční reagenty pro přenos nukleových kyselin do buněčných linií MDA-MB-231, MDA-MB-468 a T-47D.The process of RNA interference regulates the transcription and intracellular expression of genes, in which specific fragments of double-stranded RNA are involved in the induction of sequence-specific repression of gene expression of homologous mRNA. RNA interference is present in all eukaryotic organisms and is therefore considered an evolutionarily old mechanism. Today, RNA interference methods are considered to be important and at the same time fast technologies that can be used to perform functional analyzes of genes and represent an important method both in basic research and in the therapy of many diseases. The work was focused on testing the effectiveness of various transfection reagents and procedures leading to optimal conditions for silencing the desired metallothionein gene. Three commercially available transfection reagents were used for the experiments to transfer nucleic acids into the MDA-MB-231, MDA-MB-468 and T-47D cell lines.
