Interakce cytokininů a ?-glutamylového cyklu glutathionu při působení abiotických stresů u rostlin

[cze][eng] Tripeptid glutathion má klíčový význam pro toleranci stresu u rostlin. Jeho degra-dace vedoucí ke vstupu do ɣ-glutamylového cyklu glutathionu je u rostlin iniciována γ‐glutamylcyklotransferasou (GGCT). GGCT mohou zprostředkovat interakci glutathio-nu s komplexní sítí signálních drah, protože geny GGCT jsou regulovány fytohormony cytokininy a působením řady abiotických a biotických stresových faktorů. Pro studium interakce ɣ-glutamylového cyklu glutathionu s cytokininy a abiotickou zátěží byla poprvé připravena dvojná mutanta ggct2;1 ggct2;2 rostliny Arabidopsis thaliana. Ošet-ření rostlin cytokininy vedlo ke snížení obsahu glutathionu, které však nebylo u dvojné mutanty ggct2;1 ggct2;2 modulováno. Geny GGCT2;1 a GGCT2;2 rovněž nejsou vy-žadovány pro inhibici elongace kořene a charakteristické změny fenotypu spojené s působením vysokých koncentrací cytokininů. Prohledávání databází ukázalo na regu-laci genů GGCT2;1 a GGCT2;2 v odpovědi na osmotický stres, zasolení či expozici těžkých kovů jako kadmium nebo selenát. Přesto však tolerance žádného z těchto abio-tických stresů nebyla pozměněna u dvojné mutanty ggct2;1 ggct2;2. Přítomnost cy-tokininů v kultivačním médiu snížila toleranci kadmia a zvýšila toleranci selenátu, avšak ani tento účinek cytokininů nevyžaduje funkci GGCT. Studium biologické rele-vance ɣ-glutamylového cyklu glutathionu při interakci s cytokininy a abiotickým stre-sem tak vyžaduje navazující výzkum.The tripeptide glutathione is involved in numerous aspects of the plant´s life inclu-ding abiotic stress tolerance. In plants, glutathione degradation in the γ-glutamyl cycle is initiated by γ-glutamylcyclotransferase (GGCT). GGCT genes are regulated in re-sponse to cytokinin or various abiotic and biotic stressors suggesting that they are in-volved in the crosstalk between glutathione homeostasis and the complex signaling network in plants. To study the interaction of the γ-glutamyl cycle of glutathione with cytokinins and the abiotic stress response, we first generated a double mutant ggct2;1 ggct2;2 in Arabidopsis thaliana. Cytokinin treatment resulted in glutathione content decline, though this cytokinin action did not require functional GGCT2;1 and GGCT2. Similarly, the characteristic phenotypic changes associated with high cytokinin con-centrations such as inhibition of root elongation were not altered in ggct2;1 ggct2;2 double mutant. Database searching revealed differential regulation of the GGCT2;1 and GGCT2;2 genes in response to osmotic stress, salinity, and exposure to heavy me-tals such as cadmium or selenate. However, the double mutant ggct2;1 ggct2;2 exhibi-ted neither altered heavy metal nor sorbitol tolerance. Furthermore, cytokinin applica-tion decreased cadmium tolerance and enhanced selenate tolerance in a GGCT-independent manner. Taken together, double mutant ggct2;1 ggct2;2 did not provide genetic evidence that the cytosolic pathway of glutathione degradation within the γ-glutamyl cycle plays an important role in cytokinin and/or abiotic stress responses. Thus, further studies need to be conducted to reveal the biological relevance of GGCT-dependent glutathione degradation.
