Adenosin je důležitou signální molekulou v mnoha fyziologických procesech u mnoha rozličných organismů. Aberantní regulace hladiny této molekuly vede k nejrůznějším patologickým efektům, jako je například těžká imunodeficience u člověka. V minulých letech byl vytvořen genetický model pro zkoumání účinků adenosinu na octomilce (Drosophila melanogaster) a pomocí tohoto modelu byl započat rozsáhlý genetický skríning na geny, modifikující odpověď na vysokou hladinu adenosinu. V této práci jsou prezentovány dílčí úkoly v rámci tohoto skríningu, které zahrnují detailnější analýzu vybraných oblastí, které byly identifikovány tímto skríningem jako oblasti obsahující dominantní supresory fenotypu spojeného se zvýšenou hladinou adenosinu. Prvním úkolem bylo vytvoření kratší, molekulárně definované delece v rámci původně objevené větší delece. Druhým úkolem bylo otestování supresorových účinku již existující kratší delece. Třetím úkolem bylo prověření úlohy potenciálního supresoru, což zahrnovalo ověření pozic P-elementů, které tento gen zasáhly a otestování účinků těchto inzercí. Posledním úkolem pak bylo zaklonování sekvence pro tvorbu próby, která by měla být využita pro charakterizaci exprese tohoto genu Adenosine is an important signaling molecule in various physiological processes in many different organisms. Dysregulation of the level of this molecule leads to many pathological consequences as Severe Combined Immunodeficiency in humans. Recently a genetic model using fruit fly Drosophila melanogaster was established to study effects of adenosine in this organism and a genetic screen for genes modifying the effects of heightened adenosine was started. This work presents partial tasks in this screen which include detailed characterization of selected regions which were identified as dominant suppressors of the phenotype associated with heightened adenosine. First task was to produce a smaller, molecularly defined deletion in range of originally identified bigger deletion. Second task included a testing of already existing smaller deletion for the suppressing effects. In the third task, a role of potential suppressor was further analyzed which included a verification of P-element insertions in this gene and their testing for suppressing effects. In the last task, the sequence for making a probe to characterize the expression of this gene was cloned
