Mitochondrie jsou součástí většiny eukaryotických buněk, kde mezi jejich hlavní funkce patří produkce ATP pomocí systému OxPhos. Systém OxPhos se skládá z jaderně a mitochondriálně kódovaných proteinových podjednotek. Kvalita těchto podjednotek (proteinů) musí být neustále kontrolována, protože u nich může docházet k defektním změnám. Za rozpoznání a odstranění poškozených mitochondriálních proteinů jsou zodpovědné specifické mitochondriální ATP-dependentní proteázy. Doposud byly identifikovány 4 takovéto komplexy aktivní v různých mitochondriálních subkompartmentech. Proteázy i-AAA a m-AAA se vyskytují ve vnitřní mitochondriální membráně. Zatímco proteáza i-AAA je aktivní v mezimembránovém prostoru, tak m-AAA proteáza funguje na straně mitochondriálního matrix. Cílem této práce bylo pomocí RNA interference a expresních studií charakterizovat buněčnou funkci lidské proteázy YME1L, která je orthologem kvasinkové i-AAA proteázy YME1, v buněčné linii HEK293. Z našich výsledků vyplývá, že YME1L je integrální membránový protein s molekulovou hmotností mezi 600-1100 kDa, jehož karboxylový konec vyčnívá do mezimembránového prostoru. Buněčná linie se stabilně "utišenou" expresí YME1L pomocí RNA interference vykazovala akumulaci podjednotek Ndufb6 a ND1 částečně složeného komplexu I a zvýšenou stabilitu... Mitochondria are found in virtually all eukaryotic cells where their main function is the production of ATP in the oxidative phosphorylation system (OxPhos). OxPhos is build-up of both nuclear and mitochondrial encoded protein subunits. Due to the potential function threatening defects, the quality of these protein subunits is constantly under tight control by specialized proteins. The recognition and selective removal of defective mitochondrial proteins is carried out by specific mitochondrial ATP-dependent proteases. So far, four such proteolytic complexes active within distinct mitochondrial subcompartments were identified. Both i- and m-AAA protease complexes are found in the inner mitochondrial membrane. Whereas the i-AAA protease is active in the intermembrane space, the homologous m-AAA protease functions on the matrix side of the inner membrane. The aim of the present work was to characterize cellular function of the human orthologue YME1L of the yeast i-AAA protease subunit YME1 using human HEK293 cell model. We found that human YME1L is an integral membrane protein with molecular weight of approx. 600-1100 kDa, exposing the carboxy-terminal domain to intermembrane space.
