Souhrn Anorexia nervosa (AN) a bulimia nervosa (BN) jsou choroby se značnými individuálními odchylkami. Genetické pozadí hraje důležitou roli v náchylnosti ke vzniku a závažnosti onemocnění. Pro vyhodnocení vztahu mezi genetickými faktory a subtypy onemocnění jsme použili genotypizaci a zároveň analýzu vývoje vybraných klinických parametrů. Sledovali jsme skupinu 75 pacientů s AN (1. studie), 127 pacientů s AN a BN (2. studie), diagnostikovaných podle DSM-4 a MKN-10 a přispěli do souboru GWAS, který čitá 2907 pacientek s AN. Výsledky 1.studie podporují asociaci polymorfizmu -1438G/A v serotoninovém receptoru 5-HT2A s onemocněním AN a přinášejí srovnání s jinými studiemi pomocí metaanalyzy. U dalších polymorfizmů odpovědných za neurotransmisi serotoninu (serotoninový transportér 5-HTT: polymorfizmus LPR a VNTR) tato studie nastínila odlišný trend asociace LPR s onemocněním AN u české populace než ostatní publikované studie. U 5-HTT VNTR polymorfizmu nebyla pozorována žádná asociace. 2. studie se zabývala vlivem hemoxygenázy-1 (zásadní protektivní faktor při metabolickém stresu) na poruchy příjmu potravy v interakci s enviromentálními vlivy. Zkoumali jsme rizika pro AN a BN založená na genetických lokusech, odpovědných za zvýšenou náchylnost k onemocnění i přidaný efekt environmentálních rizikových stresových... Anorexia Nervosa (AN) and Bulimia Nervosa (BN) are diseases with considerable individual variation. Genetic background plays an important role in disease susceptibility and severity. To evaluate the relationship between certain genetic loci and diseases subtypes we genotyped and analysed evolution of selected clinical parameters. We investigated a group of 75 pacients with AN (1. study), 127 DSM-4 and ICD-10 diagnosed patients with AN and BN (2. study), and contributed to sample of 2907 AN patients in large GWAS study. Results from the 1st study support association of polymorphism -1438G/A in serotonine receptor 5-HT2A with AN and compare the results from other studies with metaanalyses. In next, polymorphism responsible for the serotonine neurotransmission (serotonine transporter 5-HTT, polymorphisms LPR and VNTR) the study shows different association trend of LPR with AN in Czech population compared to other studies. 5-HTT VNTR polymorphism had no observed association. The second study investigated the role of hemeoxygenase 1 (plays a pivotal role in metabolic stress protecting cells) in eating disorders, in interaction with enviromental stress.
