Do jaké míry konformační preference ukrytá v základních stavebních blocích proteinů určuje jejich trojrozměrnou strukturu? Rozsáhlé kvantové-chemické výpočty spojené s moderními solvatačními metodami představují jedinečnou sadu nástrojů k objasnění klíčových faktorů biomolekulární struktury ab initio. Na modelových systémech představujících krátké peptidové fragmenty byl provedeno úplné konformační vzorkování (sampling). Získané výsledky ukazují, jak tyto konformační preference mohou spoluurčovat tvorbu prostorové struktury proteinů. Zároveň poskytují nesmírně cenná data pro nalezení optimálního algoritmu, který účinně dosáhne pokrytí (ideálně všech) nízkoenergetických konformerů delších a delších peptidových fragmentů. Large-scale quantum chemical calculations coupled with modern solvation methods represent unique set of tools to elucidate key determinants of the biomolecular structure ab initio. Full conformational sampling was performed on model systems representing short peptide fragments. The computed data reveal some of the underlying physico-chemical principles determining the spatial structure of proteins, and provide very important data for finding and tuning the optimal algorithm that may provide a full coverage of (ideally all) low-energy conformers.
