U imunodeficientních pacientů, např. s cystickou fibrózou (CF), oportunní patogen Staphylococcus aureus způsobuje chronické infekce respiračního traktu, které se léčí dlouhodobě antibiotiky (ATB). Vystavení antibiotik ovšem může vést k navození perzistence, což se v důsledku toho projeví opakovaným návratem infekce. Cílem této práce bylo sledovat u vybraných dvojic chronologických izolátů S. aureus pocházejících z dýchacího traktu pacientů s CF, jak se v časovém období mění míra schopnosti perzistovat. Zjistila jsem, že schopnost perzistovat se v rámci klonální dvojice po dvou letech přežívání v hostiteli příliš nemění a že schopnost perzistovat záleží na adaptačních mutacích izolátů. Tvorba perzisterů může záviset na mutacích v operonech pro alternativní sigma B faktor (sigB) a hlavní regulátor virulenčních genů (agr). Duálním barvením DioC2(3) a To-pro- 3 se mi podařilo v průběhu křivky hynutí po působení antibiotik určit změnu membránového potenciálu a propustnosti membrány. Rozdělení do subpopulací dle těchto parametrů závisí především na použitém antibiotiku. Z toho vyvozuji, že různá antibiotika mohou indukovat různé mechanismy pro navození perzistentního stavu. Dále jsem pro stanovení exprese sigB a agr operonů sestavila konstrukty plazmidů se značeným promotorem (sigB-GFP, agr-GFP), avšak... In immunodeficient patients, for example with cystic fibrosis (CF), the opportunistic pathogen Staphylococcus aureus causes chronic infections of respiratory tract that are treated with antibiotics (ATB) in the long term. However, exposure to antibiotics can lead to persistence, thereby result a recurrence of infection. The aim of this work was to examine in selected pairs of S. aureus chronological isolates from the respiratory tract of CF patients how their ability to form persisters is changing in time. I have found that the ability to persist within the clonal pair does not change significantly after two years of survival in the host, and that the ability to persist depends on the adaptative mutations of the isolates. Persister formation may depend on mutations in operon of the alternative sigma B factor (sigB) and the major virulence gene regulator (agr). By dual staining with DioC2(3) and To-pro-3, I was able to determine the changes in membrane potential and membrane permeability during the killing curve with ATBs. The distribution into subpopulations according to these parameters depends primarily on the antibiotic used.
