Insulin je peptidový hormon nezbytný k udržení stálé hladiny glukózy v krvi. V lidském těle interaguje s dvěma isoformami receptoru, IR-A a IR-B. Tyto receptory se liší především tkáňovou distribucí v organismu, přičemž IR-A má spíše funkci mitogenní, kdežto IR-B metabolickou. Cílem této práce bylo vytvořit analog insulinu, který bude selektivní k IR-B. Připravili jsme tři nové analogy insulinu s triazolovým můstkem v pozicích B26 a B29. 1,2,3-triazolová skupina byla připravena cykloadiční reakcí katalyzovanou ionty Cu(I) mezi postranními řetězci kyseliny azidopentanové (N3Pent) v pozici B26 a propargylglycinu (Prg) v pozici B29. Analogy se navzájem lišily konfigurací atomů C nepřirozených aminokyselin v polohách B26 a B29. Konkrétně jsme připravili analog 1 s D-N3PentB26 a D-PrgB29, analog 2 s D-N3PentB26 a L-PrgB29 a analog 3 s L-N3PentB26 a D-PrgB29. Nové analogy byly testovány pro vazbu vůči oběma isoformám receptoru insulin (IR-A a IR-B). Analogy 1 a 2 byly oba slabší ve vazbě než lidský insulin a nevykazovaly selektivitu pro žádnou z isoforem. Analog 3 byl 4x silnější ve vazbě na IR-B a 2x silnější ve vazbě na IR-A než lidský insulin. Avšak vazebná selektivita tohoto analogu vůči IR-B (asi 2x) byla nižší než selektivita (asi 2,6x) jeho diastereoisomeru s L-N3PentB26 a L-PrgB29 připraveného v... Insulin is a peptide hormone responsible for maintaining glucose homeostasis in the circulation. Insulin interacts with two isoforms of the insulin receptor, IR-A and IR-B, which have different tissue distribution. IR-A is supposed to have rather mitogenic function and IR-B rather metabolic function. The goal of this study was to develop insulin analog, which will be more selective for IR-B than human insulin. We prepared three new insulin analogs with a 1,2,3-triazole bridge at the positions B26 and B29. The triazole bridge was formed by Cu(I)- catalysed cycloaddition between side chains of azidopentanoic acid (N3Pent) at B26 and propargylglycine (Prg) at B29. The analogs differed in configurations on C carbons of unusual amino acids at the positions B26 and B29. Specifically, we prepared insulin analog 1 with D- N3PentB26 and D-PrgB29, insulin analog 2 with D-N3PentB26 and L-PrgB29 and insulin analog 3 with L-N3PentB26 and D-PrgB29. New analogs were tested for their binding to both isoforms (IR-A and IR-B) of the insulin receptor. Analogs 1 and 2 were less potent in binding than human insulin and had no selectivity for receptor isoforms.
