S rostoucí populací a klimatickými změnami stoupá i snaha o vyšlechtění výkonnějších plodin. Genové inženýrství otevřelo nebývalé šlechtitelské možnosti ve vyvíjení rostlin s požadovanými vlastnostmi. Transgenní plodiny s lepšími vlastnostmi včetně odolnosti k nepříznivým podmínkám prostředí mohou přispět k řešení problému hladomoru a malnutrice v rozvojových zemích. Ačkoliv společnost vnímá geneticky modifikované plodiny spíše negativně, hojně se jich využívá jako krmiva pro hospodářská zvířata a mimo Evropu i pro výživu lidí. Vzhledem ke komplexní povaze odolnosti vůči abiotickým stresům se dříve nepředpokládala využitelnost genových manipulací pro šlechtění odolných rostlin. Ukázalo se však, že modifikace stresové signalizační kaskády či transkripčních faktorů může vést k úspěchu. Tato práce shrnuje možnosti genetické modifikace plodin, jež mohou vyústit v lepší toleranci vůči chladu, a je orientována zejména na rýži. Část práce se zabývá transdukcí chladového signálu, jejíž modifikací lze také docílit zvýšené tolerance vůči chladu. Další část se zabývá transkripčními faktory, jež aktivují expresi genů zvyšujících odolnost vůči nízké teplotě. Největší pozornost věnuji transkripčním faktorům CBF/DREB, jež hrají v odolnosti vůči chladu klíčovou roli. Klíčová slova: CBF/DREB, genové inženýrství,... With increasing population and climate change, there has been an increase in efforts to breed more efficient crops. Genetic engineering has opened unprecedented breeding possibilities in developing plants with desired traits. Transgenic crops with better qualities, including resistance to adverse environmental conditions, can contribute to solving problems of hunger and malnutrition in developing countries. Although society perceives genetically modified crops rather negatively, these crops are widely used as feed for livestock and outside Europe also for human nutrition. Because of the complexity of resistance to abiotic stress, the utility of genetic manipulations for the breeding of resistant plants was previously not anticipated. However, it turned out that modification of the stress signalling cascade or transcription factors can lead to success. This thesis summarizes the possibilities of genetic modification of crops, which may result in better tolerance to cold, and is mainly focused on rice. Part of the work deals with transduction of cold signal, whose modification can also result in increased tolerance to cold.
