[cze][eng] Obezita je spojena s řadou metabolických komplikací včetně inzulínové rezistence, dyslipidemie a zvýšeného krevního tlaku (metabolický syndrom). Aktivita endokanabinoidního systému (ECS) u obezity je zvýšena, čímž může docházet k dalšímu prohlubování metabolických komplikací. Farmakologická léčba založená na blokádě kanabinoidního receptoru CB1 vedla ke snížení hmotnosti a k významnému zlepšení metabolických parametrů u obézních pacientů. Nicméně paralelní ovlivnění centrálního nervového systému vedlo k nežádoucím účinkům jako je úzkost a depresivní nálady. Experimentální studie v poslední době ale naznačují, že podávání omega-3 polynenasycených mastných kyselin z mořských zdrojů (EPA, DHA) v dietě může tlumit aktivitu ECS v periferních orgánech (tuk, játra, pankreas), a tak částečně bránit metabolickým komplikacím při obezitě. Jedním z možných mechanismů účinku EPA a DHA by mohlo být vytěsňování arachidonové kyseliny z sn-2 pozice membránových fosfolipidů, čímž dojde ke snížení nabídky substrátu pro tvorbu endokanabinoidních molekul. The endocannabinoid system (ECS) activity is elevated in obesity, which can further potentiate metabolic impairments. Pharmacological treatment based on the cannabinoid receptor CB1 blockade led to a decrease in body weight and significant improvements of metabolic parameters in obese individuals. However, parallel effects on the central nervous system resulted in unwanted side-effects including anxiety and depressive moods. Recent experimental studies suggested that dietary interventions with omega-3 polyunsaturated fatty acids of marine origin (EPA, DHA) can decrease the ECS activity in peripheral tissues (adipose, liver, pancreas), and thus partially protect against metabolic disturbances in obesity. One of the underlying mechanisms behind the effects of EPA and DHA could be a replacement of arachidonic acid from the sn-2 position of membrane phospholipids, thereby reducing the substrate availability for the synthesis of endocannabinoid molecules.
