Tereza Cyrusová Školitel: Prof. RNDr. Petr Solich CSc., Prof. Angelina Pena, Ph.D. Název diplomové práce: Optimalizace HPLC metody pro stanovení polycyklických aromatických uhlovodíků v hlavonožcích Polycyklické aromatické uhlovodíky (PAU) jsou látky nebezpečné jak pro životní prostředí, tak pro zdraví člověka. Významnými deriváty PAU jsou jejich monohydroxyderiváty, které vznikají při detoxikaci PAU v těle člověka i dalších organismů a které byly označeny jako marker expozice PAU. Cílem této práce bylo optimalizovat analytickou metodu pro stanovení vybraných monohydroxyderivátů (1-hydroxypyren, 2-hydroxyfluoren, 1-hydroxynaftalen, 1-hydroxyacenaften) v tělech hlavonožců. Použitou metodou je vysokoúčinná kapalinová chromatografie s fluorescenční detekcí. Byl proveden výběr vlnových délek pro detekci, složení mobilní fáze, studie kolon, ověření možnosti použití antioxidantu, degradační studie pro jednotlivé látky a následně optimalizace stanovení všech čtyř látek zároveň. Na základě těchto kroků byla pro dílčí experimenty vybrána kolona ACE C18 a mobilní fáze ve složení metanol a voda v poměru 5:95. Monohydroxyderivatives of PAH are their very important derivatives that arise in the detoxification process of PAH in the body of human and other organisms. Some monohydroxyderivatives of PAH have been assessed as a marker of PAH exposure. The main goal of this work was to optimize the analytical method for determination of selected PAH monohydroxyderivatives (1-hydroxypyrene, 2-hydroxyfluorene, 1-hydroxynaphtalene, 1-hydroxyacenaphtene) in the bodies of cephalopods. The used method was high-performance liquid chromatography with fluorescence detection. The detection wavelengths and mobile phase composition were selected. A column study, verification of antioxidant application and degradation study were carried out.
