Závěry ► Byla popsána schopnost proteinu ARPC2, podjednotky ARP2/3 komplexu, vazby na mikrotubulární cytoskelet. Tyto výsledky společně s lokalizačními experimenty proteinů ARPC2 a ARP2 naznačily možnost, že ARP2/3 komplex může být přímým zprostředkovatelem interakce mezi mikrofilamenty a mikrotubuly v procesech spojených s 7 přestavbou mikrotubulárního cytoskeletu v koordinaci s růstem závislým na aktinu v rostlinné buňce. ► Bylo prokázáno, že vliv růstového retardantu ancymidolu na vznik tvarových malformací buněk BY-2 spočívá v jeho dosud nepopsaném inhibičním účinku na syntézu celulózy. Mikrotubulární cytoskelet v tomto procesu hrál pasivní roli a nebyl za tvarové malformace přímo zodpovědný. ► Bylo zjištěno, že během zrání somatických embryí smrku jsou buňky meristematické a buňky suspenzorové odlišně citlivé vůči latrunkulinu B, a to zřejmě díky odlišnému složení aktinových izoforem v těchto buňkách. Aplikace velmi nízkých koncentrací latrunkulinu B tak vede k selektivní smrti buněk suspenzoru a následně k selekci kvalitních a zdravých embryí. ► Toxické působení hliníku způsobuje rychlou zástavu růstu kořenů. Bylo zjištěno, že mezi její bezprostřední příčiny patří rigidizace plazmatické membrány kořenových buněk a zástava endocytózy. K časným efektům působení hliníku patří též stabilizace kortikálních... 6. Conclusions ► The capability of ARPC2 protein (subunit of ARP2/3 complex) to bind microtubules was described in this study. Our results indicated that ARP2/3 complex may mediate direct interaction between microfilaments and microtubules in processes where the coordination of microtubule reorganization and actin mediated growth was required. ► We showed that cell-malforming effect of growth retardant ancymidol was based on its inhibitory action on cellulose synthesis. Microtubules played a passive role in this process and were not directly responsible for changes of the cell shape induced by ancymidol. ► Meristematic and suspensors cells in maturating somatic embryos differed in sensitivity to latrunculin B, probably due to different composition of actin isoforms in these cells. Application of low doses of latrunculin B resulted in selective death of suspensor cells and thus contributed to the development of high-quality embryos. ► Aluminium caused rapid cessation of root growth of Arabidopsis thaliana seedlings. We showed that the immediate reason of root growth cessation was plasma membrane rigidization and loss of endocytosis in root cells.
