Imunomagnetická separace je založena na vazbě protilátky s antigenem, kde protilátka je navázána na magnetickém nosiči. V této práci byly pro imunomagnetickou separaci bakterií mléčného kvašení použity částice magnetické perlové celulózy s navázanou protilátkou antiLactobacillus a antiBifidobacterium. Metoda imunomagnetické separace byla nejprve optimalizována a následně byla ověřena účinnost a specifita navázaných protilátek na čistých bakteriálních kmenech a kvasinkách, které byly identifikovány pomocí metody polymerázové řetězové reakce. Účinnost imunomagnetické separace byla rovněž ověřena na masném probiotickém výrobku, ze kterého byly úspěšně izolovány bakterie rodu Lactobacillus. U DNA z těchto vyizolovaných buněk byla provedena vysokorozlišovací analýza křivek tání, pomocí které byla prokázána přítomnost několika kmenů bakterií rodu Lactobacillus.
Immunomagnetic separation is based on binding of antibody with antigen, where antibody is bound to magnetic particle. In this thesis there were used particles of magnetic pearl cellulose with antiLactobacillus and antiBifidobacterium antibodies. Immunomagnetic separation method was optimalized and verified for its efficiency and specifity with bacterial and yeast cells. This cells were identified by polymerase chain reaction. Efficiency of immunomagnetic separation was verified on probiotic meat product, where Lactobacillus cells were isolated. With DNA from isolated Lactobacillus cells the high resolution melting was performed. The results show presence of several bacterial strains of Lactobacillus species.
