Genetic variability in sporadic colorectal cancer: Searching for novel risk, prognostic and predictive biomarkers.

Colorectal cancer (CRC) is a major public health problem worldwide. Despite improvements in the diagnostic process and advancement in the treatment methods, the prognosis remains poor. To improve survival rates, it is important to identify people with the predisposition for CRC and to detect the potentially curable early stage of the disease. Furthermore, identifying those who would have an adverse clinical outcome associated with a particular chemotherapy would help to avoid redundant chemotherapy burden in patients and contribute to enhanced therapeutic efficacy, while minimizing treatment-related toxicity. The aim of the Thesis was to search for novel promising diagnostic, prognostic and predictive DNA-based biomarkers of sporadic form of CRC. As each patient is genetically unique, these biomarkers would aid clinicians in better diagnosis and/or in the selection of an optimal type of therapy for an individual CRC patient based on their molecular profile. In order to explore this issue, we investigated several candidate genes in healthy individuals as well as in newly diagnosed cancer patients. Analysis of prognostic features in patients with breast cancer and colorectal cancer.

Cancers represent second the most common cause of death in the Czech Republic. The most common are breast and colorectal cancers. Identification of prognostic factors improving decision-making approaches for treatment optimization belongs to the key aims of clinical research in oncology. Carriers of mutation in cancer-susceptibility genes represent a small but clinically important group of high-risk patients. The implementation of NGS have accelerated predisposing genes analyses. The large extent of data about the presence of variants in predisposing genes is in striking contrast to only a very limited information available about clinico-pathological characteristics of mutation carriers. Determination of the risk of tumor development in carriers of rare mutations or variants of unclear significance in genes with incomplete penetrance represent substantial drawbacks of current NGS analyses. To address these issues, we have attempted i) to introduce a unified approach to NGS analysis in breast cancer patients, ii) to characterize importance of prognostic factors in BRCA1/BRCA2 mutation carriers, and iii) to identify the cancer risks in carriers of germline mutations in the CHEK2 gene. Analysis of circulating markers in patients with solid tumors

Circulating cell-free DNA (cfDNA) and its tumour-derived circulating tumour DNA (ctDNA) fraction are considered an innovative prognostic and predictive biomarker in oncological diagnostics. Many studies have demonstrated higher levels of cfDNA concentration and integrity, as an indicator of the amount of ctDNA in cfDNA, in body fluids from patients with cancer diseases in comparison with healthy individuals, which suggest its potential as an effective biomarker for monitoring of the tumour dynamics. This study focused on optimisation and validation of measurement methods later used for analysis of cfDNA concentration and integrity in blood samples from patients with four different solid cancers. Two different commercial isolation kits have been tested in plasma and serum samples. Quantitative real-time polymerase reaction (qPCR) and PicoGreen dsDNA assay were optimized to effectively quantify low concentrations of cfDNA, subsequently compared to each other and to droplet digital PCR assay tested on selected samples. The concentration and integrity of cfDNA from plasma samples of breast, ovarian, colorectal and pancreatic cancer patients were evaluated. Comparison of alternative league formats in european football

Title: Comparison of alternative league formats in european football Objectives: The aim of this work is to analyse play - offs of chosen european league formats. The other aim is to make a prediction about czech league. Methods: A method of comparison was used in this thesis. A placement of each team before play - offs was compared to a placement of the same teams after play - offs. After that, there was a comparison if play - offs have more impact on championship round or on relegation round. Based on conclusions from these comparisons we have made an estimate of development in Czech league. Results: We discovered that play - offs have more impact on teams which played relegation round. In most of analysed seasons play - offs did not change the winner of the league and neither helped a lot of teams to enter European championships. We estimate that play - offs of Czech league will help more second league teams to promote to first league. Keywords: football, tournament systems, league, prediction
Assessment of Fluctuation in the Company and Suggestions for its Improvement

The master thesis analyzes the development of fluctuation associated with the labour market in the Czech Republic. By using the fluctuation data of the chosen telecommunication company in the last few years and the statistic methods, the aim is to create a prognosis of the development of fluctuation. It should result in suggestions for improving the current unsatisfactory development. The next objective is to identify the risks, which the company management have to pay attention to and which will be used to determine the corrective measures for improving the current state.

Bachelors Thesis Consequences of the hip developmental dysplasia on the musculoskeletal system in childhood and adulthood is processed in the form of case study research. Introduction of the theoretical part contains basic information about anatomy and kinesiology of healthy hip joint, its etiology and pathogenesis, options of diagnostics and treatment. In the last section of the theoretical part are described consequences of this diagnosis in patiens. The last section contains a case study of a patient with developmental dysplasia of the hip, which regularly visits rehabilitation.
Expression and prognostic value of microRNAs in patients with early breast cancer.

Breast cancer is the most frequent cancer in women and is prognostically very heterogeneous. Early breast cancer has an excellent overall prognosis with long-term survival above 90%. In this group we can also find patients with highly unfavourable progress with a risk of future disease relapse. Due to effective anticancer treatment is a main task of precise clinical decision to determine risk of an individual patient in the term of cancer relapse. We can use clinical (tumor diameter, lymph nodes) and pathological markers (grade, ER, PgR, HER2, and Ki-67), all of them have low individual sensitivity and specificity. Molecular tests based on multigene DNA or RNA assays have higher sensitivity and specificity but their interrelated concordance is low. One of the main scientific task is to find almost specific and sensitive prognostic biomarkers. microRNAs are small, highly stable, non-coding RNAs, which regulate tens of mRNAs and proteins inside cells. In cancerogenesis, they could act as oncogenes or tumor supressors as well and affect main steps of initiation and progression of cancer. The diploma thesis deals with the issue of determinig the value of the company operating in the automotive industry. The thesis deals with summarizing the theoretical backgrounds, which also explain the methods and procedures that are related to the valuation of economic subjects themselves. The analytical part identifies the company, processes the financial statements and, on the basis of them, creates the prerequisite for a correct numerical representation of the strategic and financial analysis. The valuation itself is part of the final portion, which is based on discounted cash flow and economic value added. Consequently, a summary evaluation of the achieved results for the monitored period 2012 – 2017 is produced.

The diploma thesis deals with econometric modelling and gold price forecast. A key factor is the multiple regression model and the ARIMA model. The first part of the diploma thesis contains a theoretical basis. The analytical part deals with modelling of gold market price and subsequent forecasting. Statistical and econometric verification using statistical methods play a very important role. The last part summarizes the results and makes suggestions for improvement.
Analysis of Attendance of Schools in the Town Třebíč

This bachelor thesis focuses on the analysis of demographic indicators of the town Třebíč. It analyzes the present state of preschool and school institutions in this town. Statistical methods of time series application are utilised here. The main objective is to make a prognosis of number of children entering kindergartens and primary schools depending on number of the born children. Predicted number is compared with the existing capacity of these institutions.
