Influence of aroma active compounds on flavour of fermented dairy products
Tvrdková, Denisa ; Pšenáková, Ivana (referee) ; Vítová, Eva (advisor)
The theme of this thesis is to determine the aroma active compounds in fermented dairy products. These substances significantly influence the palatability of these dairy products. Five kinds of dairy products were analyzed. Two types of creamy yogurts, and three types of yoghurt milks were produced at the dairy Ekomilk Ltd. in Frydek - Mistek and four types of acidified milk, five kinds of creamy yogurts, two kefir milks and two clabbered milks come from production of the company Kunin, Inc. Ostrava - Martinov. The SPME extraction method was used for the determination of aromatics, subsequent analysis was performed using gas chromatography. Compounds contained in fermented milk products were identified and quantified on the basis of the measured standards. Total 39 compounds, including 16 alcohols, 7 acids, 6 aldehydes, 6 ketones and 4 esters were identified. Fermented dairy products were twice subjected to sensory evaluation by the ranking test and scale test. The assessors were chosen from students, postgraduate students and staff of faculty of chemistry BUT. Consumers evaluated very positively yogurts, yogurt drinks and acidified milks, while kefir milks and clabbered milks did not suit in some aspects.
Fragrance Allergens in Foods and Everyday Use Products
Divišová, Radka ; Buňka, František (referee) ; Hojerova,, Jarmila (referee) ; Omelková, Jiřina (advisor)
Fragrances are added to food, cosmetics and other products primarily for enhancement of their sensory quality (taste and/or aroma). However, the usage of these compounds is associated with the wide range of various adverse biological effects. Some fragrances widely used in cosmetics are proved to cause e.g. skin sensitization, rashes, dermatitis, headache, cough etc. To protect the health of consumers, European union approved the cosmetic directive (ES/1223/2009) that requires the labeling of 26 allergen fragrances on the final product label, if concentration exceeds the given level (0,01 % for rinse-off and 0,001 % for leave-on product). However, some producers hide the presence of allergens in the final product under the general term „aroma“. For this reason it is very important to monitor the content of these substances in cosmetic products. Fragrances are found not only in cosmetics, but the various types of food are also aromatized by them. Unlike cosmetics, the food producers are not obliged to label the presence of fragrance allergens on the packaging. Therefore, the monitoring of these fragrances is highly desirable because of the potential health risks they pose. The aim of this study was to develop a method for simultaneous determination of regulated fragrance allergens in food and everyday use products. The method based on extraction of analytes by solid phase microextraction coupled to gas chromatography with FID detection (HS-SPME-GC-FID) was chosen on the basis of the literature review. The fiber CAR/PDMS provided the highest extraction efficiency among the SPME fibers tested. Univariate and multivariate data analysis were used to optimize the main parameters affecting microextraction process. The final method validation was performed in terms of linearity, repeatability, reproducibility, recovery, limits of detection and quantification. The optimized and validated method was applied to a wide range of products including cosmetics, aromatized food and fragranced toys. These products were also subjected to sensory evaluation especially in terms of taste and/or aroma (i.e. flavour), which may be associated with the content of the monitored fragrances. The profile test (EN ISO 13299) and seven-point category ordinal scale (ISO 4121) were used for sensory evaluation.
Assessment of aroma active compounds in currant
Lišková, Michaela ; Babák, Libor (referee) ; Vítová, Eva (advisor)
This master thesis deals with the assessment of aroma active compounds in fruits of currant (Ribes spp.). The theoretical part was focused on basic characteristics and chemical composition of currants, as well as the basic principles of techniques used: solid phase micro extraction (SPME), gas chromatography (GC) and sensory analysis. The main aim of the experimental part was to determine aroma active compounds in currants using SPME-GC-FID method. The profile and scale tests were used for assessment of sensory quality and tastiness of fruits. In total 18 varieties, among them 6 white, 6 red and 6 black currants, were evaluated. Fifty aroma active compounds were identified including 21 alcohols, 10 aldehydes, 2 acids, 8 ketones and 9 esters. The results were statistically evaluated using principal component analysis (PCA) to investigate the influence of single compounds on sensory quality of fruits.
Beer bioflavouring by dry hopping using czech cultivars of hops
Gajdušek, Martin ; Štursa, Václav (referee) ; Pořízka, Jaromír (advisor)
This diploma thesis deals with the influence of dry hopping on selected analytical and sensory properties of beer. The effect of dry hopping was observed using Czech hop varieties Kazbek and Uran, which were added to the wort during the main fermentation phase in doses of 3 and 6 gdm-3. The contact time of hops with wort was 3, 6 and 9 days. The experimental part describes the technology of preparation of the reference beer, in which dry hopping were subsequently performed. A parallel measurement was performed on each sample. In terms of the basic parameters of beer, the effect of dry hopping, especially its length, on the ethanol content was observed. With a longer period of dry hopping, the concentration of ethanol in beer increased. The higher alcohol concentration was also associated with a decrease in the apparent extract. In terms of color and pH of beer, the effect of dry hopping has not been proven. Elemental analysis performed by optical emission spectrometry with inductively coupled plasma revealed an increase in the concentration of calcium, magnesium, and iron due to dry hopping. No statistically significant difference was observed for manganese and barium compared to the reference. In terms of bitterness, a significant increase was identified in dry hopped beers compared to the reference, the value being dependent on the dose of hops used. The effect of the hop variety has not been proved. Concentrations of organic acids determined by ion-exchange chromatography with a conductivity detector were affected by dry hopping only in the case of lactic acid and acetic acid. An increase in lactic acid was observed compared to the reference, also related to the degree of fermentation. While in the acetic acid content the hop samples showed a lower concentration than the reference sample. The concentrations of myrcene, humulene and geraniol were determined by gas chromatography with a mass detector. These are volatile components of hop essential oils. Dry hopped samples recorded significantly higher concentrations of all aromatic substances compared to the reference. In terms of hop time, the highest concentrations were shown by samples hopped for three days. The results of the sensory analysis show that the overall best rated sample is a sample hopped with the Kazbek variety with a dose of 3 gdm-3 for three days. Beer which was dry hopped for the shortest time also showed the lowest intensity of bitterness, which was perceived positively.

Loupancová, Blanka ; Vávrová, Milada (referee) ; Kráčmar, Stanislav (referee) ; Fišera, Miroslav (advisor)
The production of high-quality foods requires the precise control of factors influencing their quality. The aim of this doctoral thesis was to monitor the influence of sterilization heating and storage time and temperature on selected parameters of model food matrix using suitable physical, chemical and sensory methods. Sterilized processed cheese was chosen as the model matrix. Processed cheeses are milk protein concentrates, produced by melting of mixture of natural cheeses, emulsifying salts, water and other dairy and/or non dairy ingredients. Desirable transformation of this mixture to homogenous, smooth, shiny mass with desired textural, structural, rheologic and sensory properties is caused by using high temperature (about 80-100 °C) during melting. Sterilized processed cheese is subjected to additional sterilization heating (above 100 °C) after production, significantly elongating its durability. The processed cheeses (dry matter 40 % w/w, fat in dry matter 45 % w/w) were analyzed in this work, the part of them was sterilized (117 °C 20 min). Cheeses were stored at various temperatures (cold 6 ± 2 °C, laboratory 23 ± 2 °C and elevated 40 ± 2 °C), sterilized cheeses for 2 years, non sterilized for 1 year. During storage, the samples for physical (instrumental measuring of colour and texture), chemical (assessment of fatty acids and aroma compounds) and sensory (paired preference test, ranking test and evaluation using scale) analyses were taken at regular intervals. The influence of sterilization heating and for another the influence of storage conditions on mentioned parameters were investigated. Non sterilized processed cheese kept their very good quality during all the declared durability (4 months), however, then the significant worsening of most sensory properties was observed. In consequence of sterilization heating the changes of cheese colour and texture were observed, confirmed by both sensory and instrumental assessment. The impairment of taste and aroma was also found. These changes are obvious immediately after production and differences were observed during all the durability. In the case of sterilized processed cheese, the significant impairment of single sensory properties was determined, the highest in the case of texture, taste and aroma. The results of instrumental techniques confirm conclusions from sensory evaluation. The measurements of colour and texture vindicate the formation of darker colour (increasing total difference of colour delta E*) and higher hardness (increasing Fmax) of sterilized processed cheeses, continuing during storage. The worsening of taste and aroma is in accordance with lower content of aroma active compounds and selected fatty acids.

This work deals with the evaluation of sensory quality of selected samples of bread. The samples of bread were bought in TESCO Stores a. s. ČR in Brno. Totally five samples were evaluated, two of them were rye-wheat and three of them were wheat-rye. All products were packaged and sliced. Purchased samples were subjected to sensory evaluation by the ranking test and scale tests. The assessors judged three main organoleptic properties of bread – taste, odour (aroma) and texture. The results were written down in the questionnaires and consequently graphically and statistically evaluated. The assessors which took part in sensory analysis were chosen from students of Faculty of chemistry BUT.
Analysis of active substances and biological effects of some non-tradizional cereals
Pecháček, Michal ; Vysoká, Marie (referee) ; Márová, Ivana (advisor)
This thesis deals with the characterization of active substances and biological effects of non-traditional cereals. Basic substances of these non-traditional cereals such as carbohydrates or proteins are characterized in this work. Active compounds such as antioxidants, polyphenols or -glucans are also characterized. These substances were mainly analysed by spectrophotometric methods. The theoretical part describes cereals in general. It focuses on grain morphology, cereal products and mostly on chemical composition. The summary of substances appearing in the cereal grain such as carbohydrates, proteins, vitamins, minerals etc. was processed in this part. In the practical part, the samples of the non-traditional cereals such as amaranth, sorghum, millet, kamut, buckwheat, quinoa, Job’s tears and teff were analysed. Amaranth, millet and buckwheat were also analysed in the form of flakes. Teff was analysed only in the form of flakes. Sorghum and quinoa were analysed also in their coloured variations. Firstly, the water extracts were prepared, then they were used for determination of antioxidant activity, the content of polyphenols compounds and flavonoids. The hydrolysed samples were used for determination of the carbohydrates. For other basic analysis, samples were used in the form of powder. Best results were measured for quinoa, amaranth and buckwheat. Besides basic analysis, the content of -glucans was measured. However, the content of -glucans in these non-traditional cereals was very low. Selected cereals were tested for cytotoxicity on human cells. Cytotoxicity was evaluated by using the MTT cytotoxicity test on human keratinocytes HaCaT and human caucasian colon adenocarcinoma CaCO-2. The sensory analysis was carried out in the last part of the thesis. There were tested 6 samples. From these samples were made muffins that were analysed and sensorically evaluated.
Production and evaluation of fermented dairy products
Jasimová, Zahraa ; Vítová, Eva (referee) ; Zemanová, Jana (advisor)
This bachelor thesis deals with the production of fermented dairy products and their sensory evaluation. As a model sample a kefir, more precisely, kefir milk, was chosen, which is a product made by using a combination of lactic acid bacteria and yeast (so-called kefir culture). In the theoretical part, milk cultures used for production of fermented products; as well as technology of individual fermented dairy products are characterized. Furthermore, their requirements for sensory quality and the specifics of sensory evaluation are described. In the experimental part, two suitable methods of sensory analysis were selected to compare three samples, two of them were commercial and served as a comparative samples and one was a manufactured sample. The task of the evaluators was to evaluate the color, appearance, taste, smell, consistency and overall acceptability of individual samples using a five-point scale. The model sample of a kefir milk manufactured in laboratory, was evaluated as the worst (least sensory acceptable) mainly due to the lumpy consistency, the taste and the smell reminded to some evaluators sheep's milk.
