Diploma thesis deals with relationship between the sustainability of buildings and indoor climate. The aim of the diploma thesis was to design the air conditioning system for operating theaters. In the experimental part the aim was to provide total microorganisms counts and total count of micromycetes on samples taken from filters. The work is divided into three parts: theoretical, design and experiment.
Microscopic fungi in caves and other underground shelters

This Bachelor thesis is meant as a literature search concerned with diversity of microscopic fungi in caves, prospectively in underground spaces which were made by humankind. The caves represent unique environment for living organisms which is characteristic with specific microclimatic conditions and with deficient sources of nutrients. I have concentrated my focus to micromycetes which were isolated from sediments, atmosphere, dead insect and excrements in caves. The next part of my Bachelor thesis is aimed at species Geomyces destructans which causes so called white-nose syndrome illness - geomycosis on hibernating bats. It is an actual problem which interests scientists such as mycologists, zoologists and parasitologists.

This final thesis deals with evaluation of microbial air pollution in breeding of laying hens at the university. The aim was to determine the methodology of sampling of microorganisms from the atmosphere using available techniques inspired by sampling procedures for other environments. The goal of the thesis was to identify microorganisms in collected samples with the Sanger sequencing method and to assess their potential impact on the health of the people working in such envitonment. The closing part includes a proposition of adequate measures to reduce the risk of occupational illnesses in this particular working environment.
Occurrence of mycotoxins in milk and dairy products

The aim of the bachelor thesis was to create a literary review focused on microscopic filamentous fungi (micromycetes) and the occurrence of their secondary metabolites, mycotoxins, in milk and dairy products. The first part of the work dealt with a general characteristic of micromycetes, their positive and negative effects, and with the description of three main genera - Aspergillus, Penicillium and Fusarium. The next part of the work was focused on the occurrence of mycotoxins in milk and in dairy products. The ways of contamination were described, and also the most common mycotoxins in milk and in dairy products, mainly in cheese, and some preventive measures for their elimination.

Microbial contamination of fruit teas

Teas are consumed daily by millions of people on the World, therefor it is important to care for their microbial harmlessness. The aim of this work was to conduct a reaserch in regards to general information about fruit teas, their manufacturing and possibilities of contamination by undesirable microorganisms. In this research were also described important bacteria and fungii and we focused on mycotoxins and the elimination of microorganisms from a food. Representation of microorganisms in teabags was evaluated in practical part of this research. We focused on total quantity of microorganisms, coliform bacteria, that can be pathogenic in higher representation and micromycetes. Genus structure of micromycetes was important in particular, because they can produce metabolites, mycotoxins, that can be a cause for many diseases. Analyses were conducted by two methods. Variant classified as A was consisting of transformation of the teabag contents into the physiological solution. Variant classified as B was consisting of watering of the teabag contents. During evaluation, both of these variants were compared. From the results of this work it was found out, that quantities of microorganisms were relativem small. It was discovered, that during the sample preparation by watering the sample with boiling water (variant B) the quantity of microorganisms will lower, in some cases they were not present at all. The presence of wider genera of micromycetes was also discovered. Three genera were dominant Aspergillus, Penicillum and Cladosporium. These free found of genera can be dangerous, because of mycotoxins, however micromytes in variant B were only represented by genera Cladosporium and Penicillum and in small measure.

Mykotoxiny v silážích

This bachelor thesis focuses on the topic of Mycotoxins in silages. The first part covers a brief description of mold (micromycetes) and the products of secondary metabolism -- mycotoxins, whitch can negative affect animal production and includes the impact of their effects on animal and people health. Second part of this work contains description of the individual steps silage fermentation, which may influence the occurrence of mold or mycotoxins in the final silage - date of harvest, dry matter, length of chaff, the application additives, filling and pamping silage mangers, the covering silage in relation to mycotoxins. It also deals with a short characterization of the most common forage species and with the negative effects caused by improper technological process.

In its theoretical part, the bachelor thesis deals with the whey, microorganisms and their metabolic products which negatively affect human health. The procedures for reducing or complete elimination of the risk of the microorganisms are also outlined. In the practical part, the thesis determines selected groups of microorganisms present in the whey. The measurement results are presented in the thesis' summary.
