Změny mikrobiální kontaminace vajec v průběhu skladování
Zemanová, Aneta
This masters thesis deals of microbial contamination of chicken eggs in storage. Test samples were tracked for the following microbial indicators: Total Viable Count (TVC), bacteria of the Enterobacteriaceae family, bacteria of the Enterococcus genus, psychrotrophic microorganisms, yeasts and molds. Microbial analysis was made for eggs acquired from market chains, originating from three different farming environments: enriched cages, cage-free and free range. Samples were stored in three different temperature environments: refrigerated at 5 °C for the whole storage period, stored in a thermostated box at 28 °C for the whole storage period and alternating between 5 °C and 28 °C temperature environments with one week storage periods each. The analysis of acquired test samples shows that TVC did not change significantly between different farming environments and storage temperature environments. Cage-free eggs show increasing TVC in long time storage (21 days). Cage-free and free range eggs also show higher count for Enterobacteriaceae. Enterobacteriaceae counts show significant differences between samples stored at constant 28 °C and alternating temperatures. Lowest microorganism count for bacteria of the Enterobacteriaceae family was observed at alternating temperatures. Long time storage resulted in elevated Enterobacteriaceae count in cage-free eggs. Presence of Enterococcus, psychotrophic microorganism and molds was detected in a low number of samples.
Evolution of sperm morphology in birds
Zemanová, Aneta ; Stopka, Pavel (advisor) ; Frolíková, Michaela (referee)
The sperm is one of the most variable cells that carry various morphological differences across species and is subject to selective pressures that may drive changes in sperm morphology. Different morphological adaptations that emerged may enhance successful fertilization. This thesis focuses on the analysis of sperm morphology in different species of songbirds, the representation of proteins in both sperm and seminal fluid, and their classification into different categories of gene ontology. The main aim of my thesis was to determine whether differences in proteomes follow the level of sperm competition or rather their phylogeny. Six species of songbirds were selected for this study: Marsh Warbler (Acrocephalus palustris), White-throated dipper (Cinclus cinclus), European greenfinch (Chloris chloris), Barn swallow (Hirundo rustica), Common chiffchaff (Phylloscopus collybita) and Zebra finch (Taeniopygia guttata). Sperm morphometric measurements were made in all the species, with the greatest variability recorded in the central part of the sperm flagellum. Using mass spectrometry, were analysed sperm and seminal fluid proteins separately and discriminant analysis was performed to determine the proteins that most differentiated the selected songbird species from each other. Obtained data provide...
Exercise motivation for 4th and 5th graders, using mobile application
Zemanová, Aneta ; Přibyl, Ivan (advisor) ; Rotková, Hana (referee)
5 ABSTRACT This diploma thesis deals with exercise motivation for 4th and 5th graders, using mobile application The theoretical part of the thesis deals with physical ability of children about 10 years old. It describes motor development of younger school-age children. It focuses on the specifics of school maturity and children's needs. It points out consequences of lack of physical activity. It deals with motivation. It researches leisure activities for children and organizations involved in them. In the practical part, the thesis uses survey to answer question. How much free time children spend using mobile phone and what kind of application they use. It offers list of application with a methodology of use and overlapping into the school environment. The list of applications serves for teachers, educators or parents, as a guide to active leisure activities with children using modern technology. Individual applications have been tested within one class. The result is a comparison of the individual applications among themselves and the suitability or effectiveness of their use. Furthermore, the text examines the influence of cigarettes to adolescents and most distinctive life situations that may affect adolescent smoking. In the practical part of the work is presented a qualitative research based on ethnographic method. In the research was worked with a group of adolescents between 15 and 18 years. Observation and semi-structured interviews was used to map the course of the smoking careers of the group members and reveal a direct social influences that may...
Studium dráhy pro syntézu mastných kyselin u klíštěte \kur{Ixodes ricinus}
ZEMANOVÁ, Aneta
Aim of this work was to study fatty acid synthesis pathway in the tick Ixodes ricinus. I have characterized tissues and stage expression profiles of three genes; acc (acetyl coenzyme A carboxylase), fas (fatty acid synthetase) and fabp (fatty acid binding protein) by qRT-PCR. Then expression of these genes was silenced by RNA interference and phenotypes observed during and after feeding in nymphs and adults. The pathway seems to be crutial for the ticks.
Ritual elements in the practices of Herbalife Independent Distributors
Zemanová, Aneta ; Šanderová, Jadwiga (advisor) ; Grygar, Jakub (referee)
The diploma thesis called "Ritual elements in the practices of Herbalife Independent Distributors" is focused on the manifestations of Randall Collins' interaction rituals on the example of multi-level marketing company - Herbalife. It tries to answer two questions: answer two questions: What does the Herbalife Independent Distributor community hold together? What motivates them to work, when everything is voluntary at Herbalife and they do not have a competitive driving force there? The connecting mechanism of the multi-level marketing structure and interaction rituals are practices of organizational culture. The most important are the physical meetings of members in which interaction rituals take place. By living interaction rituals, Herbalife members produce emotional energy, by which they empower the symbols of organization and the sense of group solidarity. Positive emotional energy has an impact on the cooperation of distributors with the organization, and it affects also their effort to achieve common goals.
