Alena Vondráčková The atorvastatin effects on endoglin expression in the atherosclerotic plaques. Diploma thesis Charles University in Prague, Faculty of Pharmacy in Hradec Králové Pharmacy Background: We observed the expression of endoglin in atherosclerotic plaques in ApoE / LDL receptor-deficient mice with regard to the administration of atorvastatin. Methods: We used C57BL/6J female mice with double deficiency of apolipoprotein E and LDL receptor at the age of 8 weeks. Animals were divided into two groups and both groups were fed special diets for 8 weeks. Biochemical analysis of blood samples was performed. Histological staining with oil red we have taken for the determination of lipids in atherosclerotic lesions. Was also performed immunohistochemical analysis of tissue taken from segments that formed the aorta together with the upper half of the heart. Detection of expression of the endoglin was performed using Avidin-Biotin method (ABC) with detection using DAB. Results: Biochemical analysis showed an increase in total cholesterol after an eight- week administration of atorvastatin. Histological staining with oil red demonstrated the presence of atherosclerotic lesions in both groups of animals. Administration of atorvastatin resulted in hyperglycemic effect and significant reductions in...
The Endoglin Expression after Administration of Spirulina Platensis in Mouse Model of Atherosclerosis.

Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Biological and Medical Sciences Candidate Mgr. Alena Vondráčková Consultant Doc. PharmDr. Petr Nachtigal, PhD. Title of Thesis The endoglin expression after administration of Spirulina platensis in mouse model of atherosclerosis. The aim of this study was to clarify whether the administration of Spirulina platensis affects the expression of endoglin and the production of collagen in atherosclerotic plaques. We also study the effect of Spirulina on blood lipid levels. We used the mouse strain C57BL/6J deficient apolipoprotein E in the age of 3 months. These mice were randomly divided into two groups. The control group was fed with atherogenic diet containing 1% cholesterol. The second group received the same diet enriched with Spirulina platensis 20 mg daily. After an eight-week experiment, biochemical analysis of blood was performed. Detection of collagen was performed by histological staining green trichrom. Imunohistochemical analysis of tissue samples of segments (aorta and upper half part of heart) were used to determine the endoglin expression. In addition, we performed imunohistochemimistry and histology quantative analysis by stereological metods. Administration of Spirulina resulted in a decrease in LDL...
