Functional analysis of syntaxin 16 phosphorylation using yeast as a model

4 Abstract Mechanism of fusion of intracellular membranes in eukaryotic cells involves several protein families including soluble N-ethylmaleimide-sensitive-factor attachment protein receptor (SNARE) proteins and Sec1/Munc-18 related proteins (SM proteins). It is known that the transport is evolutionary conserved from yeast to man. Therefore for facilitating of the research, we can use simple eukaryotes Saccharomyces cerevisiae. Mammalian SNARE protein syntaxin 16 has a yeast homologue Tlg2p which is used in this study as a model for studying affects of phosphorylation to the syntaxin 16 function. Also their binding partners, SM proteins mVps45p (mammalian) and yeast Vps45p are homologous. Phosphorylation of SNARE proteins is known as a possible way of regulation of membrane fusion. Abolishment of one of the putative phosphorylation sites in Tlg2p protein, serine 90 leads to dominant effects on the exocytic and endocytic pathways. The work presented in this study shows some phenotypes of mutants based on this phosphorylation site of protein Tlg2p. Those mutants are S90A (cannot be phosphorylated) and S90D (phosphomimetic - acid carboxyl group mimics phosphate group). Case study of a patient with a primary diagnosis of ankylosing spondylitis

Title: Case study of a patient with a primary diagnosis of ankylosing spondylitis Objectives: The aim is to become familiar with the general issues of ankylosing spondylitis and in a special section to develop a case report of physical therapy care for the patient with a diagnosis of ankylosing spondylitis. Methods: Bachelor thesis includes a case study of a patient with a primary diagnosis of ankylosing spondylitis. The work consists of a general part, which contains a brief description of the disease and possible complications of diagnostic and therapeutic methods. In the second part - special patient case report is processed, which was created during my professional experience in Rheumatology Institute in the period from 8.2.2013 to 14.1.2013. In addressing this diagnosis was not used any invasive techniques. Results: Performed after therapy there was a significant reduction in pain and restore proper movement stereotypes. AfD's support in federal states of Bremen, Saxony-Anhalt and Baden-Württemberg

This thesis concerns the support for the AfD in the federal states Bremen, Saxony-Anhalt, and Baden-Württemberg. In particular, the goal is to explain the support for the AfD among the eligible voters of Bremen, Saxony-Anhalt, and Baden-Württemberg at the most recently elections held in Bremen on 10 May 2015 and in Baden-Württemberg and Saxony-Anhalt on 13 March 2016. This is a disciplined, interpretive case study, which uses the existing models of voting behaviour to explain the voting for the AfD in Bremen, Saxony-Anhalt, and Baden- Württemberg. It employs a sociological model of voting behaviour and a model of rational choice. The goal is to determine whether the support for the AfD in the three selected federal states can be explained using these models of voting behaviour, and if so, which model of voting behaviour best explains the support for the AfD. First, the thesis presents the models of voting behaviour that are used in the study for analysis of the support for the AfD. The next portion of the thesis deals with the party Alternative for Germany. The section of the thesis that follows is devoted to the actual analysis of support for the AfD, first in Baden- Württemberg and then in the federal states Saxony-Anhalt and Bremen. The position of the governments of Gerhard Schröder on Turkey's accession to the European Union

The following thesis deals with the pro-Turkish policy of red-green coalition in Germany during 1998-2005, which represented a turning point in German Turkish policy, because it did not follow the anti-Turkish policy of Helmut Kohl. Also today it represents an unique policy - nor the current Chancellor Angela Merkel initially followed it, whose image of Europe is incompatible with the membership of Turkey in the European Union. Based on an analysis of the most important debates held in the Bundestag under the red- green coalition government, where was discussed the Turkish question, the thesis tries to answer the question why the government of Gerhard Schroeder/Joseph Fischer supported Turkey's accession to the EU. It concludes that in the speech acts of individual government officials and MPs of the governing parties intermingled both strategic arguments and arguments corresponding with the classic postwar milieu shaping strategy that aims to influence the milieu beyond the borders of the national state. Strategic arguments gained in importance due to three international events that shook the world during the governments of Schroeder/Fischer and which tightened the active Turkish policy. Invasive alien plants in Trebonsko region

The Bachelor's thesis deals with the issue of invasive plant species. Following invasive plant species were mapped in the South Bohemian region of Třeboň: Canadian goldenrod (Solidago canadensis), Giant goldenrod (Solidago gigantea), Bohemian knotweed (Reynoutria x bohemica), Japanese knotweed (Reynoutria japonica), Sakhalin knotweed (Reynoutria sachalinensis) and Himalayan balsam (Impatiens glandulifera). The goal of this thesis was to map the above-mentioned species in one part of Stropnice Site of Community Importance and its surroundings. Furthermore, the thesis also aimed to record the most frequently colonized biotopes according to the invaded locations. The results show that Canadian goldenrod colonized seventeen locations and therefore it is one of the most invasive species. Next, Himalayan balsam was found in seven locations and Japanese knotweed in five locations. Last, Bohemian knotweed and giant goldenrod each colonized only one location. In this area, such species most often invade biotopes affected by people and locations near water resources. The plants also grow mostly in wet localities.
