Short Stories Milan Jariš' (Oni přijdou) and E. F. Burian's (Osm odtamtud): Theirs Versions
Vlková, Kateřina ; Holý, Jiří (advisor) ; Špirit, Michael (referee)
This master's thesis focuses on two collections of short stories that have not been analyzed enough so far, which deal with the period of Nazi occupation and persecution. Both are based on the authors' own experiences: They Will Come (orig. Oni přijdou; first published in 1948) by Milan Jariš and Eight Stories from Over There (orig. Osm odtamtud; first published in 1954) by E. F. Burian. The main attention will be focused on the textual comparison of the different versions of these titles (for Jariš, new versions in 1949, 1953, 1956 and 1985; for Burian, new version in 1956). The theoretical starting points will be the studies of Oldřich Králík and Miroslav Červenka examining different variants and versions of specific literary works.
Employment Relationship in Terms of Wage Costs Optimalization
Vlková, Kateřina ; Růžičková, Diana (referee) ; Musilová, Helena (advisor)
This bachelor’s thesis addresses the labour relations in terms of optimizing labour costs. The theoretical part defines the basic concepts related to labour relations from the legal, tax and economic point of view. Proposed solutions that could lead to labour costs optimization were based on an analysis of the employer’s current situation in the area of labour relations.
Photochemical internalization of saporine by phthalocyanine photosensitisers
Vlková, Kateřina ; Macháček, Miloslav (advisor) ; Kozlíková, Magdaléna (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Student: Bc. Kateřina Vlková Supervisor: RNDr. Miloslav Macháček, Ph.D. Consultant: Mgr. Monika Rohlíčková Title of the diploma thesis: Photochemical internalization of saporine by phthalocyanine photosensitisers The application of therapeutic macromolecules is often affected by limited cellular uptake and lysosomal degradation, thus failing to reach their intracellular targets. Their cellular uptake can be increased through photochemical internalization. Photochemical internalization uses photosensitive agents that localize in the membranes of endosomes and induce drug release upon exposure to light. This diploma thesis is focused on the evaluation of the effect of saporin in the combination with non-symmetrical photosensitizers on HeLa cell line using photochemical internalization. Effect of various concentrations of these compounds on the viability of tumour cells was evaluated. Two approaches to deliver the energy from light were compared as well: before and after the administration of saporin (i.e., "light-before" and "light-after" approach). From obtained data, it is evident that saporin (administered alone) does not significantly reduce the viability of HeLa cells, even though it was applied in a...

Genomic analysis of Paenibacillus larvae and its relation to virulence
Vlková, Kateřina ; Hrabák, Jaroslav (advisor) ; Fišer, Radovan (referee)
Paenibacillus larvae is a Gram-positive sporulating bacterium that causes American foulbrood (AFB). It is one of the most dangerous bacterial pathogens of the honeybee (Apis mellifera). P. larvae spores are highly infectious to bee larvae and resist physicochemical influences. P. larvae is subtyped using repPCR with ERIC primers (Enterobacterial Repetitive Integrance Consensus) into five genotypes (ERIC I-V), which possess different colony morphology, metabolism and especially virulence. There is a significant genetic variability among isolates of P. larvae, which may contribute to differences in virulence. P. larvae isolates used in this work were obtained from clinical cases of American foulbrood as well as from a debris collected from bee hives with no American foulbrood symptoms from all over the Czech Republic in cooperation with the Beekeeping Research Institute, s.r.o., Dol. The isolates were obtained from larvae and hive debris. Both virulet and avirulet strains were sequenced using the SMRT (single molecule real time) method on the Sequel platform (PacBio). This method is suitable for Whole Genome Sequencing (WGS), because it allows sequencing of long reads with high accuracy, eliminating the effect of a large number of repetitive sequences during the genome assembly. Furthermore,...
Role of uric acid in the pathogenesis of cardiovascular diseases
Vlková, Kateřina ; Kovařík, Miroslav (advisor) ; Kudláčková, Zděnka (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biological and Medical Sciences Student: Kateřina Vlková Supervisor: PharmDr. Miroslav Kovařík, PhD. Title of the bachelor thesis: The role of uric acid in the pathogenesis of cardiovascular diseases The basic biological function of uric acid in the human body is to protect cells from oxidative damage. More precisely, its behavior depends on the physico-chemical environment in which it is located. Under certain conditions and higher concentration in serum, it may act as a prooxidant and participate in the pathophysiology of a variety of diseases. These are mainly kidney disease, non-alcoholic steatosis of the liver, diabetes mellitus II. type and metabolic syndrome. These diseases can precede cardiovascular risk. Cardiovascular failure may be caused by some mechanisms. These mechanisms include especially oxidative stress, inflammation, reduced availability of nitric oxide and endothelial dysfunction, proliferation of smooth muscle cells in blood vessels, insulin resistance or metabolic disorders. The cause of cardiovascular disease associated with uric acid has not been proved. It can only serve as a intermediator of harmful effects on vascular tissue and myocardium. Keywords: uric acid, oxidative stress, inflammation,...

Vlková, Kateřina ; Holý, Jiří (advisor) ; Merhaut, Luboš (referee)
This bachelor's thesis focuses on short stories of Ladislav Fuks that were published in the first decades of his literary activity. Next to the printed books of stories (Mí černovlasí bratři published in 1964, completed already in 1958; Smrt morčete, 1969, proses written between 1953 and 1968) the work focuses on short stories published in magazines (Květen, Věstník židovských náboženských obcí, Židovská ročenka) and short stories deposited in the Museum of Czech Literature where the author's manuscripts can be found. The aim of the thesis is to observe the ontogenesis of Fuks' short stories and his poetics. KEYWORDS: Ladislav Fuks, Short Stories, 1950s, 1960s
