TOWNHOUSE ON THE WATERFRONT IN PŘEROV

House with a day care for seniors on the waterfront in Prerov. It is a three-storey building with parking on the ground floor. There is also a medical attendance, a cafe and a hairdresser´s on the ground floor. On the first and the second floor there are apartments for seniors. The design of the apartments corresponds with needs of handicapped people.
Znojmo – Capuchin monastery and town walls moat

The main topic of this thesis is reconstruction of the monastery like a nursing house for elderly people. The main reasons for this solution is very advantageous position of the monastery in the historic center, the possibility of using the existing building and possibility to use gardens of monastery like an area for relaxation. In gardens of monastery is designed new house for elderly people. This new building is designed with the greatest emphasis on tolerance to the surrounding environment and the historical importance of this area. The footprint of the house follows the trace of the original fortification, which led to the toe of Vlčí věž. It´s floor plan arrangement recalls the letter U. in the center of the house, there is an inner courtyard, which corresponds with the inner courtyard of the monastery. The design of new object is trying to find the best position to fit into the context of the town of Znojmo. At the same time, this arrangement allows the solution to the inner garden, resting place of relaxation, which refers us back to the monastery garden, which occurred in these places since the time of establishment of the monastery.
Low-rise Timber House

The Bachelor thesis deals with a design of timber building. The object is a two-storied cellarless building with flat single-leaf roof which is designed for stay of four people. KLH timber boards with external insulation layers form the main construction system of the building. The thesis includes contract documents for the construction of building and seminar thesis (paper) that elaborates on the isssue of timber buildings.

Children's Home, Novy Jicin

The Master´s thesis deals with a design of the children´s home in Novy Jicin. It is an object providing preventative educational assistance to children, pupils and students and their families, schools, and school establishment. The institute will operate as an outpatient (advisory panel) and boarding (forward determined length of stay based on the decision of the social employee, e.g. two months). The house is two-storied with partial basement and flat roof. The supporting structure of underground floor is made up of concrete blocks, reinforced concrete ceiling. Supporting structures above ground floors including ceiling are made up of solid timber boards. The roof is double-leaf, structure is made up of timber nailed trusses. The building is founded on the concrete base strips. Thermal insulation is made from cannabis plates. The thesis includes contract documents for the construction of building.

The main subject of the thesis "Communication strategy of company" is analyzing the current state of the communication mix, based of the analysis result and marketing re-search to design a suitable communication mix. The thesis is divided into three main parts. The first part focuses on the theoretical interpretation and definition of important marketing concepts, communication mix, marketing environment and marketing re-search. The second part introduces basic information about reconditioning a studio and analyses the external and internal environment and based on marketing research the suit-able segment is chosen. The last part of the thesis evaluates the current state of the communication mix and based on the results of the second part of the thesis a new communication mix of reconditioning studio Figuraline is proposed.
Analysis of genomic variants in patients with nerodevelopmental diseases with a focus on epilepsy.

Epilepsy is a chronic neurological disease affecting millions of people worlwide. The etiology of epilepsy is heterogenous. Published studies confirm a strong genetic basis. While the genetic causes play a huge role in the development of epilepsy, the etiology remains unclear in many patiens. We present a cohort of 400 patients with epilepsy, who underwent whole genome testing focused on the detection of copy number variants (CNVs). The main criterion for inclusion into the group was manifestation of epilepsy in isolated form or in combination with other neurodevelopmental disorder or congenital anomalies. Genome-wide analysis was performed using two different platforms of array CGH (array comparative genome hybridization): SurePrint G3 CGH ISCA platform 4x180K and 8x60K (Agilent Technologies). For evaluation of clinical impact of detected CNVs different databases (DGV, ClinVar, OMIM, DECIPHER) and relevant articles were used. In our cohort we have detected 2730 CNVs in total and 86 of them (detected in 76 individuals), were evaluated as possibly clinically relevant - 82 CNVs were evaluated as a possible cause of epilepsy and 4 of them were evaluated as secondary finding without relationship to the patients' phenotype. In my work I deal with methods of sensory evaluation of grapevines. In the first part I research informations about structure of grapevines, as well biochemistry of grapevines and as well factors affecting of maturity of berry. In the second half of work I identify informations about types of maturity berry, and about sensory and analytical evaluation. In experimental part of work I do sensory evaluation directly in the vineyard and analytical evaluation in the laboratory. For evaluation I have varieties as Pinot Blanc, Sauvignon Blanc, Hibernal, Savilon and Bronner.
The mechanism of impaired formation of head-tail coupling apparatus in centrobin deficient acephalic spermatozoa

Acephalic sperm syndrome (ASS) is a rare form of teratozoospermia that is probably genetic in origin. The sperm of individuals with this syndrome have a damaged apparatus connecting the head and flagellum (HTCA), which leads to the formation of acephalic sperm, i.e. sperm with separate heads from flagella. Individuals affected by ASS are almost exclusively infertile. One of the proteins whose mutation causes ASS is SUN5. Together with KASH proteins, SUN proteins are part of the so-called LINC complex, which ensures the connection of the nucleoskeleton with the cytoskeleton. Centrobin is a protein involved in centriole duplication and assembly of the dividing spindle. Rats carrying a defective gene for centrobin have reduced fertility and exhibit an ASS phenotype. For that reason, this model organism was used in this work to study the possible interrelationships of proteins involved in the formation of ASS. Given the similar phenotype of centrobin and SUN5 mutated spermatozoa, there is a possibility that centrobin interacts with one of the LINC complex proteins and together they participate in the formation of the head-flagella junction.
