Translation reinitiation mechanism on mRNA of trascriptional activator GCN4.

Translation reinitiation is a gene-specific translational control mechanism exploiting the ability of some short upstream open reading frames (uORFs) to retain post-termination 40S ribosomal subunit on the mRNA. Reinitiation efficiency depends on cis-acting sequences surrounding the uORF, translation elongation rates on the uORF, selected initiation factors, and the intercistronic distance of the short uORF from the main ORF. Although the precise mechanism of reinitiation is still not known, great progress in elucidating some of its details has been recently made with help of the GCN4 translational control model system. Among them, involvement of eIF3 was shown to play a critical role for efficiency of this process. In particular, it was proposed that eIF3 specifically interacts with sequences located upstream of a reinitiation-permissive uORF upon termination, and that this step is instrumental in stabilizing the 40S ribosomal subunit on the mRNA to allow subsequent resumption of scanning for reinitiation downstream. In this thesis, the current knowledge of the translation reinitiation mechanism is summarized. As a typical example, the yeast transcriptional activator GCN4 has been chosen, the mRNA of which is subjected to a tight translational control via the very reinitiation mechanism.
Respect to human dignity of critically ill patients at ICU and ARD

This bachelor's thesis is focused on the issue of respect for dignity of patients at Intensive Care Unit (ICU) and Anaesthesiology and Resuscitation Department (ARD). The main objective of this work was to determine how nurses perceive and respect the human dignity of patients in intensive care. Furthermore, to determine which procedures are the most harmful to dignity and whether nurses respect the rights of patients and the Code of Ethics. The theoretical part is divided into three chapters. The first chapter defines the basic terms: human dignity, intimacy, shame, personality, morality, ethics and patients' rights. The second chapter is focused on document Essence of Care 2010, part of which deals with the issue of respect for human dignity in health and social care. The third chapter describes the important virtues, qualities and communication skills of nurses. The practical part describes objectives, methodology and organization of research. The research group involved 86 nurses (10 men, 76 women). All respondents work at ICU and ARD at the General University Hospital in Prague. The role of the N-terminal domain of the a/TIF32 subunit of eIF3 in mRNA recruitment to the 43S pre-initiation complexes.

Translation initiation is a complex process which results in the assembly of the elongation competent 80S ribosome from the 40S and 60S ribosomal subunits, the initiator tRNA and mRNA, and is orchestrated by numerous eukaryotic initiation factors (eIFs). Although it represents one of the most regulated processes of gene expression, the exact mechanism of one of the key steps of translation initiation - mRNA recruitment to the 43S pre-initiation complex (PIC) - is still only poorly understood. Recent studies indicated that besides eIF4F and poly(A)-binding protein, also eIF3 might play an important, if not crucial, role in this step. In our laboratory, we recently identified a 10 Ala substitution (Box37) in the a/TIF32 subunit of Saccharomyces cerevisiae eIF3, which interfered with translation initiation rates. Detailed analysis showed that this mutation significantly reduces the amounts of model mRNA in the gradient fractions containing 48S PICs as the only detectable effect in vivo. Moreover, a recently solved crystal structure of the N-terminal part of a/TIF32 pointed to two Box37 residues, Arg363 and Lys364, both proposed to contribute to one of the positive, potentially RNA-binding areas on the a/TIF32 surface. The reflecting of Folkways in Nursery School education Plan

In my bachelor thesis I point to the possibility that render traditions in the creation and implementation of educational content of school curriculum kindergarten. I suppose that the program based on folk tradition can fully meet requirements for pre-school education in the curriculum for pre-school education. In the theoretical part I focus on the folk traditions that are natural and connected with nature. I´m trying to integrate these traditions into pre-school education. I describe folk traditions and their traces in my region. In the practical part I present the vitality of folk traditions in nursery school. These are activities and actions that can be carried out in nursery school. The content of school curriculum has been compiled using folks traditions. All these topics are added with thematic unit Feasts. The thematic unit is accompanied by a video as a cross-sectional image.
