Diagnostic markers specific language impairment from the perspective of a speech therapist
Veselá, Šárka ; Klenková, Jiřina (advisor) ; Kopečný, Petr (referee)
This thesis deals with specific language impairment. The theoretical part introduces related terminology, classification, etiology, symptomatology and diagnostic issues. The research section analyzes the different linguistic levels in a sample of children and devotes a common specific variations in verbal production that could be designed as diagnostic markers. The qualitative study was an analysis of verbal production of nine children ranging in age from 5.4 to 5.7 years, with specific language impairment. In all the statements were reflected deficits in the use of sibilance, skipping voice in the first position, disruption of understanding to varying extents, incorrect naming and difficulties výbavností words and disturbance of speech and other prosodic factors. Morphology and syntax has always seen the most significant disruption, but was very heterogeneous. Powered by TCPDF (www.tcpdf.org)
Flow cytometry in the diagnostics and characterization of congenital disorders of glycosylation
Veselá, Šárka ; Hansíková, Hana (advisor) ; Hodek, Petr (referee)
Congenital disorders of glycosylation (CDG) are rare multisystem metabolic diseases and their number has rapidly grown in recent years. The clinical manifestation includes very broad spectrum of symptoms. In most of all cases CDG are caused by mutations in genes encoding the enzymes of glycosylation pathway. Based on the type of defect, CDG are divided into the following groups: disorders of N-glycosylation or O-glycosylation of proteins, defects in modification of proteins by GPI anchor, disorders of lipid glycosylation and defects that impact multiple glycosylation pathways. The aim of the thesis was to find new biochemical analyses suitable for diagnostics and characterization of CDG patients. The experimental conditions were optimized for selected markers (Sambucus Nigra (SNA) lectin, proaerolysin (FLAER), antibodies to proteins CD55 and CD59) and the staining was applied to cultivated skin fibroblasts from controls and patients diagnosed with CDG by whole-exome sequencing (ATP6AP1-CDG, PIGN-CDG, SLC10A7-CDG, PISD deficiency). The experiments were performed using flow cytometry (FACS) and fluorescent microscopy (FM). The detection of sialylation by SNA lectin and analysis of the mitochondrial membrane potential changes by a fluorescent labelled probe JC-1 with FCCP simulation of mitochondrial...
Interaction of selected flavonoids with N-acetyltransferases
Veselá, Šárka ; Hodek, Petr (advisor) ; Bořek Dohalská, Lucie (referee)
Flavonoids are natural compounds synthesized in plants as secondary metabolites. Due to broad range of their biological effects, for example we can mention antioxidant, hepatoprotective or antibacterial effects, flavonoids are increasingly used as a component in dietary supplements. Besides positive effects, some negative effects were observed, especially mutagenic and pro-oxidative properities. The biotransformation is an important process which is affected by flavonoids. Interactions between flavonoids and biotransformation enzymes in both phases of biotransformation are essential for the metabolism of drugs and carcinogenesis processes. In this bachelor thesis the influence of selected flavonoid compounds, myricetin and dihydromyricetin, on the activity of N-acetyltransferase 1 and 2, enzymes of second phase of biotransformation, was studied. Mainly the mechanism of this influence was tested. The inhibition of human recombinant and of N-acetyltransferase 1 and 2 by myricetin and dihydromyricetin was proved. The more in-depth study of interactions of N-acetyltransferase 1 and 2 with selected flavonoids shows that raising concentration of second substrate (p-aminobenzoic acid, sulfamethazine) does not lead to reduction of inhibition effect. Thus, it is highly probable that both flavonoids already...

School maturity
Veselá, Šárka ; Zemková, Jaroslava (referee) ; Felcmanová, Lenka (referee)
This thesis deals with the issue of school readiness and procedures for the assessment. It focuses on achieving individual developmental areas for children before they enter school, the conduct of the assessment of maturity and option diagnostic tools. The objective of this work is to identify most vulnerable and high-risk areas for school children before entering school and the possibility of stimulation. It also presents the possibility of using screening tests and presentation of their results. The aim of this thesis is based on the utilization of available screening tests in a sample of children, including the latest diagnostic tools maters, the practical part of the work and the conclusions of this thesis, which dealt with the appropriate options stimulation attenuated developmental areas. Gender analysis of the novel 1984 or where lies the boundary of Orwell's analysis of power
Veselá, Šárka ; Knotková - Čapková, Blanka (advisor) ; Nováková, Soňa (referee)
My master's thesis entitled Gender analysis of the novel 1984 or where lies the boundary of Orwell's analysis of power focuses on the literary representations of masculinity and femininity. The core of this analysis lies in an interpretative analytical work focuses on the uncovering of the ideological gender representations. The work addresses the issues of how the fictional reality is genderized, which unreflected ideological messages are t ransmitted to the reader, to which patterns are the reader interpellated by the text and whether an uncritical attitude towards misogeny appeals to common sense of the reader and whether this attitude is the other aspects of the classical novel. The theoretical section of this thesis is inspired (drawn, based) mainly by Anglo-American feminist literary criticism from the 1970s and 1980s and poststructuralist and postcolonial theory. However, the character of the work is quite interdisciplinary, this section utilizes many methodological and theoretical concepts of which the most important are concept of resisting reading and the Foucault concept of power. The analytical section concentrates on how masculinity is utilized as a framework of analysis of the concept of games and games imagery which allows for the differentiation of the structure of an antiutopic...

The French-speaking childish literature translated into Czech
VESELÁ, Šárka
This bachelor's degree dissertation speaks about the French-speaking childish literature translated into Czech. The theoretical part presents the childish literature in general terms, its functions, features, divisions and some types and forms. It also deals with the relationship of the precocious child with the book and the importance of translations in the childish literature. The next chapter expounds in detail the kamishibai traditional Japanese method which is an original storytelling way and an unusual manner to approach a literary work. The practical part consists of the choice of French-speaking childish books that have been translated into Czech and their presentation. The goal is to put them forward not only to the nursery school teachers but also to the parents and readers.

Possibilities and limits of preschool education of french language based on the example of the international preschool Blue Sky
VESELÁ, Šárka
This diploma thesis is focused on the topic of preschool education of french language, its possibilities and limits. The question is considered from a psychological perspective and the starting point is the mental evolution of child. The main aim is to discover if it?s possible to teach french language in the early years, from three to six years old, and, if so, how and with which results. Investigations were conducted in two preschools for which the author prepared and with a native speaker taught french lessons. Final results of this thesis follow from this personal experience of the author.
