The effects of Glycopyrronium Bromide and Tiotropium Bromide on tight junctional proteins of blood-brain barrier mimicking cells and their transport across a blood-brain barrier in vitro model
Vaněček, Václav ; Štaud, František (advisor) ; Čečková, Martina (referee)
In this thesis, we present information on the effect of glycopyrronium and tiotropium on tight junctions in the blood-brain barrier. We also studied transport of these substances through an in vitro model of blood-brain barrier. Using ELISA method, we confirmed the effect of glycopyrronium on the expression of ocludin, ICAM-1 and claudine-1 and the effect of tiotropium on the expression of claudine-1. By transport studies across the ECV304 cell monolayers, we conclude that both substances have the potential to migrate through the blood-brain barrier.
Nanotechnology and biomaterials for application in cell therapy of spinal cord injury
Vaněček, Václav ; Haninec, Pavel (referee) ; Smetana, Karel (referee)
New approaches for the treatment of SCI use advances in the fields of nanotechnology, biomaterial science and cell therapy. The results presented in this thesis showed that superparamagnetic iron oxide nanoparticles coated with a stable dopamine-hyaluronane associate can be used for the safe and effective labeling of MSC. Cell labeling efficiency, viability and the relaxivity of the tested particles were significantly better than those obtained with the commercial particles Endorem®. The DPA-HA coated nanoparticles can be used for the noninvasive monitoring of cell therapy using MRI. Furthermore, we showed that SPION can be used for the targeted delivery of MSC to the site of a spinal cord lesion. The labeled cells can be concentrated in the lesion area by means of a magnetic implant. The process of cell targeting depends on the physical characteristics of the magnetic implant as well as on the biological features of the cells and nanoparticles, as we described with a proposed mathematical model. It is possible to modify the properties of the magnetic system, e.g. by changing the shape or size of the magnet, thus tuning the magnetic force distribution and the gradient of the magnetic field necessary for effective cell targeting. A promising therapeutic strategy for the treatment of spinal cord injury is the...
Vybrané aspekty hospodářského rozvoje Čínské lidové republiky po roce 2000 a její 13. pětiletý plán 2016-2020
VANĚČEK, Václav
The Diploma thesis "Selected economical growth aspects of the People's Republic of China after year 2000 and 13th five-year plan 2016-2020", aims to describe economical development after the year 2000 and to pinpoint important aspects of the economical growth. As the key source for this thesis were used mainly English written sources and internet statistical portals. For source processing and data evaluation were used heuristic and comparation method. The first part of this work is devoted to recent economical history as well as to description of the economical situation of PRC at the turn of the millennium. Macroeconomic indicators analysis afthe the year 2000 follows next. As the data source for this part serves OECD, The World Bank and also the PRC National Bureau of Statistic portals. The next part of this thesis describes important aspects of China's economical development in the recent history. The main focus is on China's direct foreign investments and their imortance for economical growth. Last, but not least, the chapter 13th five-year plan focuses on the new strategies and policies such as are "Made in China 2025 or The New Silk Road. This chapter also uncovers the senitive questions around PRC's "green economy" and social climate changes.
Injury of professional athletes - soccer players, rehabilitation and prevention
Vaněček, Václav ; Tuček, Milan (advisor) ; Šulcová, Margaréta (referee) ; Bartoš, Miroslav (referee)
The study analyses available data (a total of 564 reports) on injuries to the musculoskeletal apparatus in a series of 240 professional soccer players, with the average age of 24.3 years as of the beginning of all football seasons (median 24.0 years, range 16 - 39 years), covering the period of 16 years (1997 - 2013). The analysed variables included age, height, body weight, BMI, different football positional roles (defence, offence, goalkeeper), number of injuries (injury to the ankle, the knee, lower limb muscles; lower limb fracture, injury to the upper limb, injury to the head, neck and back, injury to tendons and groins) and the surface on which the injury was sustained (dry - hard, wet - soft, artificial). Processing of statistical data and examination of a potential correlation between the respective injury and variables was based on the mixed model method. During the season the risk of a knee injury sustained in a match was statistically significantly higher as compared to training; at the same time the risk was decreasing with the growing BMI value. In addition to a statistically significant training/match variable (with matches posing a higher risk), the risk of injury to the knee depends on the type of surface, where dry surface is associated with a higher risk of such injury. Matches,...

Selected economic and social indicators of the People's Republic of China in the last period. Basic trends
VANĚČEK, Václav
The Bachelor's degree thesis, "Selected economic and social indicators of the People's Republic of China for the last period. Basic trends", aims to describe the economic and social situation of PRC during the last period and to capture the underlying economic trends. As the key source for this thesis was used the publication by Ronald Coase, How China Became Capitalist. The first part of this work is devoted to the beginnings of the socialist market economy and the character of Deng Xiao Ping. This part mainly focuses on the reform period since the early 70s. Another part deals with the analysis of the key macroeconomic indicators and comparing official statistics of the Chinese Statistical Office with the data from the internet portals such as The World Bank or OECD. The last part of the thesis is devoted to other frequently mentioned topics such as China's position in the BRICS group, the last five-year plan or changes in the social climate during the last period.
