Characterization of individual phases of cultivation of red yeasts

This bachelor thesis deals with the problematic of metabolic activity and morphology of several biotechnologically significant red yeasts - Rhodotorula glutinis, Sporobolomyces shibatanus and Phaffia rhodozyma during the cultivation time. These yeasts were cultivated on glucose medium, during the cultivation samples were taken for the analysis of produced metabolites. The amount of -carotene, astaxanthin, coenzyme Q and ergosterol were monitored. The amount of biosynthesised lipids and fatty acid profile was also investigated. HPLC/PDA and GC/FID were used for the analysis of these metabolites. Yeasts were cultivated 56 hours (they reached early stationary phase). The highest production of carotenoid pigments showed Phaffia rhodozyma (1856.58 g/g), 42 % of this amount represents -carotene. The highest lipids biosynthesis efficiency was reached by Rhodotorula glutinis. To study the inner structure of yeast cells the Fluorescence Life-time Imaging was used, the Autofluorescence of carotenoids and fluorescence of lipid bodies induced by lipophilic dye Nile red was observed. This method is very useful for imaging cell inner structure (membranes, some kinds of organelles). The changes that the cell undergoes during its evolution could be quantified too. The relationship between the level of carotenoid concentration, membrane rigidity and metabolic shift was found.

Tato práce se zabývá strukturní studií dvou proteinů: proteinu Tail morphogenetic protein H (TmpH) bakteriofága 812, který napadá Zlatého stafylokoka (Staphylococcus aureus) a hexamerinu z potemníka (Tribolium castaneum). S. aureus je jedním z nejvíce rezistentních patogenů způsobující onemocnění s vysokou morbiditou a mortalitou. Bakteriofág 812 je schopen infikovat a lyzovat 95 % kmenů S. aureus a má potenciální využití ve fágové terapii. Protein TmpH je součástí virionu tohoto fága. V rámci této práce bylo připraveno několik plazmidů nesoucích gen TmpH, které byly použity pro rekombinantní expresi proteinu v buňkách E. coli BL21(DE3). Protein byl vyčištěn afinitní a gelovou chromatografií. Pro čistý protein byly optimalizovány krystalizační podmínky. Hexamerin je nejhojnějším proteinem larev a kukel hmyzu s dokonalou proměnou. V průběhu metamorfózy hexamerin slouží jako zdroj aminokyselin. V rámci této práce byl hexamerin izolován z kukel potemníka T. castaneum. Pro stanovení struktury hexamerinu byly použity dvě metody: rentgenová krystalografie a kryo-elektronová mikroskopie. Byly optimalizovány podmínky pro růst krystalů a vypěstovány krystaly vhodné pro sběr difrakčních dat. Nicméně struktura hexamerinu byla rychleji vyřešena kryo-elektronovou mikroskopií s rozlišením 3.2 . Znalost struktury hexamerinu umožní pochopení jeho funkce v regulaci vývoje hmyzu s dokonalou proměnou.
Knowledge of patients with type 2 diabetes mellitus about nutrition and physical activity

This diploma thesis is focused on the knowledge of patients suffering from type 2 diabetes mellitus. The main aim of this work is to determine how well these patients are educated about type 2 diabetes mellitus depending on both the duration of the disease and whether they attend a nutritional therapist. The work is divided into a theoretical and a practical part. The theoretical part provides general information about diabetes mellitus. There is a marginal description of pancreatic function and the history, definition and classification of diabetes mellitus. Type 2 diabetes mellitus, its pathogenesis, clinical picture, diagnosis, screening, therapy and associated complications, both acute and chronic, are discussed in more detail. As part of type 2 diabetes therapy there is the most discussed non-pharmacological treatment, which includes dietary measures and physical activity. The practical part was formulated on the basis of quantitative research, which was carried out using a questionnaire survey. Data were collected from November 2019 to March 2020. 150 questionnaires were distributed to different inpatient departments and clinics in Jičín Hospital. 106 completed questionnaires were returned, but 24 of them had to be rejected due to incomplete or incorrect completion. Alimentation of hypercholesterolemic patients

The Bachelor thesis is focused on eating of patients suffering from hypercholesterolemia. The main goal of Bachelor thesis was to find out how patients with hypercholesterolemia eat and what impact to the cholesterol level does the change of eating habits have. The thesis is divided into theoretical and practical part. The theoretical part describes the basic food ingredients, cholesterol and characteristics of hypercholesterolemia, its types, risk factors influencing its development, treatment options and prevention of the illness. It deals with the influence of individual food ingredients to cholesterol level and diet measures to decrease cholesterol level in blood. In conclusion the theoretical part describes atherosclerosis, which is closely related to hypercholesterolemia. The practical part was created by a qualitative research using semi-structured interviews. Interviews were lead with six patients suffering from hypercholesterolemia with high concentration of cholesterol in blood. Then the patients were instructed to record their menus for one week. I entered these menus into "Nutriservis Professional" program, which helped me evaluate them and thanks to this I was able to determine the average cholesterol intake in a patient's diet. Then I gave the patients a diet recommendation they should follow for the next six months. After this period, patients were asked to check their cholesterol level again, which helped me to find out the influence of the diet to level of cholesterol. It can be seen from the results of the research that in case the diet is kept, and a physical activity is included into a normal life, the patients can successfully decrease cholesterol level in blood. We can notice that most patients exceeded the recommended 300mg daily dose of cholesterol. The Bachelor thesis can serve as a teaching material for patients suffering from hypercholesterolemia, but it can also serve to increase knowledge about this disease.
Marketing Communication of Green Ways Company

The bachelor thesis is focused on the marketing communication of the Green Ways company. The aim of the thesis is to assess the current marketing communication and based on questionnaire research appraise, whether it is executed efficiently. The research shows that the company´s marketing communication is relatively effective, imperfections are rather seen at salespeople of the Green Ways company. The thesis might be contribution to the aforementioned salespeople, who have the opportunity to increase the efficiency of their work, based on the results of the research.

The Life-Course of Volunteers

This bachelor thesis analyses the life course of volunteers who engage in voluntary work in hospitals. The research sample comprises five volunteers four women and one man within the age range of 40 to 55. The objective of the study was addressed using a qualitative research methodology involving a narrative interview. The theoretical part of this paper discusses the concept of a life course, its development and new approaches to this concept and the inter-relations between the course of one's life and volunteering. It also examines volunteerism in general as well as volunteering in hospitals together with the activities it involves, and gives an overview of volunteers' motivations. The practical part presents research results. The main purpose of the research was to analyse the life course of volunteers. Partial objectives were to examine the relationship between the course of volunteers' lives and their volunteer work and the impact of volunteering on their lives. The respondents gave account of their history (family background, education, career path and personal situation, free-time activities). The research results indicate that while the life course of individual volunteers differs, some similarities and overlap can be found. Also, despite that the links between one's life path and volunteering were perceived differently by each volunteer, certain similar patterns (psychological predispositions and personality traits, having achieved a certain age, difficult life events, the need to do something meaningful) were also observed. The same applies when exploring how volunteering affected the lives of volunteers. The results of the research may benefit coordinators of hospital volunteer programmes and other interested parties. Further research could focus on volunteers involved in other areas of volunteering, or a similar study could cover a larger research sample.

The topic of my thesis is Innovation website Spa and Convention Hotel Thermal in Karlovy Vary. The topic I chose because of the ever-increasing demands on information technology and computer technology. Tourists are increasingly demanding, and it is increasingly difficult to meet their needs and fulfill their requirements. To any company could best present, must have created the best website. In the analytical part focuses on the characteristics of the location and the hotel itself and create a SWOT analysis of the current website. In the practical part, based on the results of questionnaires created, I designed a presentation that corresponds to the identified requirements.
