Possibilities of hematopoietic stem cells determination in the peripheral bood.

In this study the hematopoietic stem cell, its properties, sources, utilization and possibilities of its determination in the peripheral blood are described. The detection of CD34+ cells by flow cytometry is the reference method for determining the number of hematopoietic stem cells. The aim of my work was to verify whether it is possible to identify such cells can also use another method. This method is the determination of hematopoietic progenitor cells by a haematology analyser Sysmex XE 2100. In the practical part of the study, data of patients obtained from the analyzer Sysmex XE 2100 and flow cytometer are processed. This is the number of hematopoietic progenitor cells, CD34+ cells and total number of leukocytes. The data are compared by using the Spearman correlation test. It has been shown that the numbers of hematopoietic progenitor cells and CD34+ cell counts, there is dependence. Correlation coefficients of 0.55 and 0.58 can be considered very well. The analyzer Sysmex XE 2100 can be used as an alternative method to determine the number of hematopoietic stem cells in peripheral blood and leukoconcentrate. Confrontation of results of coagulating arguments measured by varios methods

Prothrombin test is the most frequently required test of hemostatic imbalances and it is also used to monitor anticoagulation treatment by coumarin preparations, e.g. warfarin. It is a basic routinely used group coagulation test that describes the so called outer conversion method of prothrombin to thrombin. Owing to the the highly frequent usage of this examination, a vast number of reagent producers appears in the market related to this testing. Reagents of individual producers vary in quality, stability, sensitivity and price. In this work I compare results of prothrombin test measured by reagents of 3 different producers (Diagon, Diagnostica Stago and Grifols) and my goal is to establish if the gained results are comparable. I used the prothrombin test with randomly chosen healthy persons and with patients of Faculty Hospital Hradec Králové. I tested each sample simultaneously with reagents of the 3 producers mentioned above. When I statistically analysed the results, I found that values measured by reagents of all 3 producers correlate quite well and are comparable to one another. Consequently, all reagents can be used without problems in clinical laboratories.
Systemic and Local Allergic Insect Sting Reaction From The Epidemiological, Diagnostic a Therapeutic Point of View

Hymenoptera venom allergy threatens the patients with severe, often life threating anaphylactic sting reaction. Proper diagnostics and treatment are therefore crucial for the patient with this diagnosis and should be performed by an experienced allergist. We aimed to provide a comprehensive look at these patients, from the epidemiological, diagnostic and therapeutic point of view. Initially, we focused on predictors of severe systemic sting reactions. Then we followed by evaluation of the utility of available laboratory diagnostic methods and by monitoring of immunological parameters during venom immunotherapy. We met all these goals. We detected higher age, sting localization in the head region and male gender as predictors of severe systemic sting reaction. We also evaluated the clinical utility of all diagnostics methods, and we even suggested the possible way, how to optimize the diagnostic procedure in patients with Hymenoptera venom allergy. We noticed significant changes in immunological parameters during venom immunotherapy. Due to a low number of stings during treatment, we have not been able to evaluate the clinical relevance of these changes yet.

Political and legal aspects in selected provisions of the Constitutional Charter of 1920

The subject of this rigorous work is to investigate the political and legal aspects that influenced and lead to the formulation of the particular provisions in the Constitutional Charter of 1920. The work also offers juristic analysis of selected constitutional provisions in the context and, in some cases, in the form of comparison with previous laws and standards that implemented the provisions of the Constitutional Charter. We also focus on those constitutional laws, which have become a part of or amendments to the constitution.

A comparative view of Czech and English prepositions signifying place and time

This thesis explores the meaning and distribution of spatiotemporal preposition in English in comparison with the Czech prepositions of the same kind. The main part of this work focuses on the usage of the preposition at that,next to on and in, is used to express both temporal and spatial ideas (e.g., at the corner, at 7:30). Investigating the meanings associated with the spatiotemporal particle at used in the gathered material may bring fundamental insights into the relation between language, mental representation and human experience. The thesis aims to identify as large amount of sequences as possible with prepositional function of at, as both languages use different ways of interpreting the usage of the preposition. Whether the English and Czech spatial and temporal usage of the at-phrase is distinct from one another in meaning and distribution; it will be discussed at greater length in the presented collection of examples.
