X-ray computed tomography of cartilage tissue of embryos

Research of the cartilage is standardly realized by optical microscopy on histological slices. However, for better understanding of shape-organizing processes, imaging in 3D is required. This thesis deals with X-ray computed tomography of mouse embryos. Specifically, it focuses on visualization of craniofacial cartilage. The thesis summarizes the basic principles of X-ray computed tomography. Short research study dealing with staining of soft tissues for X-ray is also provided. The procedure of implementation of the tomographic measurement of mouse embryos stained by PTA is described. This procedure includes the staining protocol, fixing the sample and setting of the CT machine. Subsequent segmentation and creating of a 3D model of cartilage is also desribed. Results for every step of this procedure are demonstrated at the mouse embryos of developmental stage from 12.5 to 17.5 days. In order to improve data quality and standardization of the whole process, parameters of X-ray tube and temperature conditions during the tomographic measurement are explored. Finally, resulting models of craniofacial cartilage are shown with the possibility of subsequent analysis.

Implementation of industrial X-ray computed microtomography in developmental biology

Rentgenová počítačová mikrotomografie (CT) je metoda pro trojrozměrné (3D) zobrazování vnitřní struktury objektů. Laboratoř rentgenové počítačové mikro a nanotomografie na CEITEC VUT (Středoevropský technologický institut, Vysoké učení technické v Brně) se zaměřuje zejména na nedestruktivní analýzu v průmyslovém odvětví. Ukázalo se ale, že tato metoda nachází široké uplatnění i v dalších vědeckých oblastech včetně vývojové biologie. Tato práce poskytuje propojení mezi materiálovými a živými vědami. Kompletní proces vedoucí k vytvoření 3D modelu různých anatomických struktur je detailně popsán. Tento postup zahrnuje kontrastování vzorků, samotné CT měření a analýzu dat. Právě zpracování dat je mnohdy považováno za nejdůležitější část v rentgenové počítačové tomografii a proto je této oblasti věnována v práci největší část. Úpravou měřicích parametrů byl dále optimalizován CT přístroj GE v|tome|x L 240 za účelem umožnění automatické segmentace. Navíc je diskutována možnost měření na synchrotronu za účelem zvýšení prostorového rozlišení a diferenciálního kontrastu. Předpokládá se, že v blízké budoucnosti získají průmyslové CT přístroje podobné zobrazovací vlastnosti, a to jak díky nedávnému vývoji v oblasti hardwaru, tak v oblasti zpracování dat.

Correlation of Thermoanalytical Data with Primary Properties of Humified Substrata

Tato dizertační práce je napsána jako termoanalytická studie humifikovaných substrátů, tj. huminových látek a půdních vzorků, získaných z několika různých zdrojů tak, aby byl pokryt co nejširší rozsah primárních charakteristik. Práce hledá lineární korelace (s využitím Pearsonova korelačního koeficientu) mezi kinetikou termo-oxidační degradace (sledovanou pomocí termogravimetrie (TGA) a vyjádřenou konverzním časem stanoveným neizotermickou izokonverzní metodou) a standardními charakteristikami, jako je množství aromatického a karboxylového uhlíku přítomného v huminových látkách stanoveného spektroskopií nukleární magnetické rezonance (NMR). Hlavním cílem této práce je objasnit proces termo-oxidační degradace humifikované části přírodní organické hmoty a hledat závislosti mezi primární a sekundární strukturou huminových látek. Dalším úkolem je nalezení korelace mezi termogravimetrickým úbytkem hmotnosti půdních vzorků a jejich mikrobiální stabilitou měřenou v laboratorních experimentech jakožto půdní respirace (tedy uvolňování oxidu uhličitého). Účelem je zjistit, zdali vzájemné propojení vůbec existuje, a objasnit jaké druhy procesů, které provázejí půdní respiraci, je možné jednoduše odhalit pomocí TGA. Výsledky práce skýtají detailnější pohled na termickou analýzu půdních vzorků a jiných humifikovaných materiálů. Hlavním přínosem této práce je prezentace vyskytujících se aplikací termické analýzy pro charakterizaci půdní organické hmoty a dále také příspěvek k rozvoji aplikací technik termické analýzy v půdní vědě.

The main effect of this graduation theses is detailed project for control process of exactness of angular measuring instruments. It is necessary to produce conponents with sufficient accuracy for correct function of machines. That is why there is very important thing to exactly measure proportions of these conponents during industrial process. Then the real parameters of finished products are confronted with measures stated in conponent plans. The final deviations have to fall into fixed limits. Předložená diplomová práce je zaměřena na aplikaci konceptů značky sloužící k rozvoji konkurenceschopnosti produktu instituce vysokoškolského vzdělávání, konkrétně studijního oboru “Entrepreneurship and Small Business Development”. Na základě teoretických poznatků, metodiky dle Ko Floora a konceptu Brand Prismu je zhodnocena stávající situace studijního oboru. Zároveň jsou provedeny analýzy konkurentů a požadavků zákazníků, společně s analýzou odvětví k určení pozice značky studijního oboru na trhu. Na základě výsledků ze všech provedených analýz jsou následně stanoveny návrhy řešení konceptu značky na podporu rozvoje konkurenceschopnosti studijního oboru.

Possibilities of preventing the negative influence of cognitive biases on everyday decision making

This bachelor thesis deals with possibilities of preventing the influence of cognitive biases on our everyday decision making, especially with the negative effect of the anchoring-and-adjustment heuristic on consumer behavior. The first chapter is concerned with human irrationality in decision making, primarily with cognitive biases. Some of the most significant types of cognitive biases that negatively affect our decision making are presented and various approaches trying to explain their origin and nature are introduced subsequently. It continues with a chapter on possibilities of the prevention of cognitive biases where the theory of debiasing is mentioned and an example of a successful debiasing strategy is presented. The last chapter of the theoretical part focuses in detail on one particular type of cognitive bias that arises from using the anchoring-and-adjustment heuristic. The aim of the empirical part of the thesis is then to propose a strategy that will, at least partially, help reducing the negative impact of this bias on our decision making in everyday situations - particularly from the consumer perspective. Population mobility in the circular economy

The aim of the thesis is to evaluate the state of mobility of the population in everyday commuting to work and school at the level of regions of the Czech Republic in the aspect of circular economy. Secondary data mainly from the yearbooks of the Ministry of Transport and Tourism, the Czech Statistical Office, the Czech Hydrometeorological Institute, etc. were used for the research. The professional assumption of circular economy in the transport issue was drawn from the documents of the European Union and the Ministry of Environment of the Czech Republic. It is on the basis of the EU documents that an "ideal region" that meets the EU requirements for circularity in transport was drawn up for the purposes of the analysis. From the above sources, the data was analysed using statistical methods and a "mobility circularity degree indicator" was compiled for the purpose of this thesis. The indicator describes and evaluates the level of mobility in each region of the Czech Republic and its result is evaluated. Finally, the findings are followed by proposals for measures to maintain or improve the state of mobility in the regions.

Using of verbal assessment by primary school teachers after distance learning

The diploma thesis deals with the use of verbal assessment by primary school teachers before and after distance learning. It is primarily focused on finding out whether and how distance learning has influenced the use of verbal assessment by primary school teachers. The theoretical part of the thesis is concerned with an explanation of the basic terms of the topic, in particular: evaluation, school evaluation, school evaluation in the context of legal standards, verbal evaluation, materials for verbal evaluation, legislation of the verbal evaluation, distance learning, recommendations for distance learning and reference for assessment during distance learning. In the practical part, an analysis of three research studies, which were carried out using the methods of two consecutive questionnaire surveys, was implemented. One questionnaire survey was carried out in 2020 and the other in 2023. In addition, interview methods were used with teachers of the primary school first grade. The surveys were aimed at obtaining the necessary data to analyse the use of verbal assessment by first-grade teachers before and after distance learning. Molecular basis of deficit of F1Fo-ATP synthase and its impact on energy metabolism of a cell

Mitochondria's primary function is to produce energy through the process of oxidative phosphorylation. ATP synthase is a macromolecular rotary machine located in the inner mitochondrial membrane that catalyzes the synthesis of adenosine triphosphate (ATP) from adenosine diphosphate (ADP) and inorganic phosphate (Pi). The mitochondrial disorders due to ATP synthase deficiency represent a heterogeneous group of diseases characterized by variable severity of the phenotype with onset at birth or later in life till adulthood. Mutations in both, mitochondrial or nucelar DNA encoded genes, may result in ATP synthase impairment, either isolated or combined with deficits of other complexes of oxidative phosphorylation. The aims of the thesis were to characterize TMEM70 protein, an ATP synthase assembly factor, and to analyze the impact of novel disease variants leading to ATP synthase deficiency in patients' derived samples. TMEM70 is a 21 kDa hairpin structure protein localized in the inner mitochondrial membrane, with both termini oriented into the matrix, which forms higher oligomer structures. Our results demonstrated that the absence of TMEM70 protein leads to an isolated deficiency of complex V followed in some stage by adaptive/compensatory effect of respiratory chain complexes.
