Svobodová, Božena ; Čihař, Martin (advisor) ; Görner, Tomáš (referee)
My thesis deals with evaluation of visits in central parts of Giant Mountains National Park in the summer month through three years of periodic monitoring and unique evaluation survey of visitors of natural protected area of Beskydy. It compares results of this protected areas each other and with other chosen national parks in North America, Australia and Europe. It has been recorded the quantitative load of selected localities in Giant Mountain National Park (Luční bouda, Labská bouda and U čtyř pánů) and natural protected area Beskydy (Pustevny). It were conducted guided interview with visitors at this locations. With this method used was obteined high ratio response (about 92 %). It was recorded a total of 44 281 visitors. The most frequented location was Luční bouda and U čtyř pánů in Giant Mountain National Park. But on the top of the Pustevny, the amount of walkers was twice times bigger. Statistically, it was subsequently found some answers to the dependence of demografic characteristic of population of respondents. From this conclusions were drawn both valid conclusions especially for the population of visitors, compared to other protected areas were also expressed about the conclusions of the participants travel mountain protected areas at all. These conclusions of the thesis will be...

Svobodová, Barbora ; Nachtigal, Petr (advisor) ; Němečková, Ivana (referee)
Barbora Svobodová Sunitinib effects on endothelial expression of hemeoxygenase-I in normotensive rats. Bachelor thesis Charles University in Prague, Faculty of Pharmacy in Hradec Králové Medical Laboratory Technician Background: The aim of this bachelor thesis was to define and describe the expression of hemeoxygenase-1(HO-1) in the aortas of normotensive Wistar Kyoto (WKY) rats and evaluate possible effects of sunitinib treatment by means of immunohistochemical and stereological analysis. Methods: Two groups of WKY rats were used for the experiment (6 animals in each of them). Sunitinib was administered to the first group. The other one (the control one) received water only. Application of sunitinib proceeded by following scheme: 8 weeks of using/ 5 weeks rest period/ 2 weeks of using. After that we performed the immunohistochemical analysis on 60 preparations of rat's aortas which observed the expression of HO-1 by methodology En Vision with DAB detection. Then the stereological evaluation was performed. Results: The immunohistochemical analysis showed the expression of HO-1 in the endothelium only and it was noticed just in several animals especially from the group with sunitinib treatment. Conclusion: The immunohistochemical and stereological analysis demonstrated a significant increase of the...
Effect of gene optimization on recombinant expression of human cytochrome P450 3A4
Svobodová, Barbora ; Martínek, Václav (advisor) ; Bořek Dohalská, Lucie (referee)
Cytochrome P450 3A4 is integral membrane protein residing in endoplasmic reticular membrane. In human the highest concentration cytochrome P450 3A4 is expressed in liver, where it plays a major role in metabolism of many drugs and xenobiotics. The main aim of the thesis was to evaluate the effect of gene optimization on heterologous expression of human cytochrome P450 3A4. At first expression constructs based on vectors pET22b were prepared. Then the efficiency of heterologous expression of optimized vs. natural gene sequence encoding truncated form of the human cytochrome P450 3A4 compared. The results show that the gen sequence optimized for E. coli strains K12 was expressed in significantly higher efficiency than the original human gene based on cDNA sequence. Another aim was to evaluate the suitability of pET22b based expression vectors for recombinant production of native (complete) form of human cytochrome P450 3A4. The amount of native form of the protein found in bacterial membrane was however substantially lower then that of the truncated form. Keywords: cytochrome P450 3A4, heterologous expression, pET22b, gene synthesis
Impact of English on the Modern Hindi
Svobodová, Blanka ; Vacek, Jaroslav (referee) ; Kostič, Svetislav (advisor)
The aim of this thesis was to provide a description of the linguistic behaviour and repertoire of the speakers of Hindi from the so-called middle-class of the capital city of Delhi, who come into contact with native speakers of English in their employment and everyday life. Therefore, the thesis starts with CHAPTER ONE, where I described and explained the methodology and material used (1.1.0), provided a definition of the middle class in India and its determinants (1.2.0.) in view to my respondents and thirdly gave a detailed insight into the multicultural and multilingual situation in Delhi (1.3.0.), where all of the respondents live. In CHAPTER TWO, I demonstrated the evolution of the language presently known as Hindi, i.e. a language that has been influenced and enriched by various cultures and languages, both from the internal sources such as the Sanskrit based words tatsama (2.1.1.), Hindi based words tadbhava (2.1.2.) and vernacular words called desi (2.2.) as well as external sources, such as Arabic (2.3.1.), Turkish (2.3.2.), Urdu (2.3.3.) or Persian (2.3.4.) and especially the linguistic influences connected with the European Colonial Powers, such as Portuguese (2.4.1.) and above all English (2.4.2.), whose influence started to be felt from the second half of the 19th century and culminated in the...

For 7 of which this is the first report of antimicrobial activity. Abuta grandifolia, Maytenus macrocarpa, Naucleopsis glabra and Pterocarpus rohrii inhibiting all tested microbial strains at MICs ranging from 0.25 to 8 mg/ml; 0.125 to 0.25 mg/ml; 0.0625 to 4 mg/ml and 0.25 to 16 mg/ml, respectively, were considered as potentially prospective sources of new bioactive substances.
The English influence on Hindī
Svobodová, Blanka ; Vacek, Jaroslav (referee) ; Kostič, Svetislav (advisor)
Hindi is the official language of India and a predominant language in the so-called Hindi belt, i.e. the states and territories of Himachal Pradesh, Delhi, Haryana, Uttar Pradesh, Madhya Pradesh, Bihar, Uttaranchal, Jharkhand, Rajasthan and Chattisgarh. Outside these areas, Hindi is widely spoken in cities such as Mumbai, Hyderabad or Kolkatta as well. Furthermore, according to the 1998 survey it is the second most spoken language in the world, with 333 million native speakers, i.e. 40 % of the Indian population. 1 Besides, Hindi is used by another 300 million lndians as their second language and by another 8 million speakers outside of India. Thus this number includes both the countries where Hindi is the common second language, such as Mauritius, Fiji, Trinidad, Guyana or Surinam and the countries, where Hindi is the first language of large Indian communities settled all over the world, such as South Africa, Yemen, Uganda, Northern America, Great Britain, Australia, Germany etc. Concerning the linguistic point of view, Hindi belongs to a vast family of lndo-European languages, particularly to its lndo-Aryan branch. lt is a descendent of Sanskrit, the earliest speech of the Aryan conquerors who settled in the north-west frontiers of India around 2000 BC. The history of Sanskrit dates more than three...
