The role of membrane cholesterol in delta-opioid receptor signaling Correlation with plasma membrane structure
Brejchová, Jana ; Svoboda, Petr (advisor) ; Novotný, Jiří (referee) ; Teisinger, Jan (referee)
Study of HEK293 cells stably expressing fusion protein between delta opioid receptor (δ-OR) and pertussis toxin-insensitive mutant of Gi1α protein, δ-OR-Gi1α (Cys351 -Ile351 ), provided the following results. Decrease of plasma membrane cholesterol content (cholesterol depletion) induced by cyclic oligosaccharide β-cyclodextrin did not affect binding of specific δ-OR agonist, [3 H]DADLE. Neither the maximum number of binding sites nor the affinity of [3 H]DADLE binding was changed by cholesterol depletion. However, the ability of δ-OR to activate cognate trimeric G proteins was impaired. EC50 value of agonist-stimulated [35 S]GTPγS binding was an order of magnitude higher. This effect was observed in case of both control and pertussis toxin-treated cells. It means that cholesterol depletion markedly reduced the efficiency of functional coupling of δ-OR to endogenously expressed pertussis toxin-sensitive G proteins of Gi/Go family as well as covalently bound Gi1α (Cys351 -Ile351 ) protein. Unchanged plasma membrane cholesterol content is therefore important requirement for proper δ-OR function. Detection of the effect of cholesterol depletion on the functional activity of δ-OR was supported by the analysis of changes in biophysical state of plasma membrane using fluorescent membrane probes,...
Power of municipalities and regions
Havlíková, Hana ; Kopecký, Martin (advisor) ; Svoboda, Petr (referee)
This dissertation's intention is to deal with the legal demarcation of regions' and municipalities' jurisdiction, with short history excurs from year 1848 to the present. Next intention are individual components of the individual and delegated jurisdiction, regulatory activity and content statutory authorization to authority to legislate and its limits. Continue to be paid to comparison of czech and European legislation contained in the European Charter of Local Self- Government. The aim of this thesis is to compare the definition of regions and municipalities, of the individual or of delegated powers, and highlight some of the differences in this definition.
Remonstrance Proceedings
Adamec, Martin ; Prášková, Helena (advisor) ; Svoboda, Petr (referee)
- Remonstrance Proceedings The thesis is focused on the remonstrance and the following special administrative proceedings. The remosntrance represents an ordinary appeal, which can be used to challenge the first instance decision that has not already become enforceable and has been issued by the central administrative office, by the minister or by the director of other central administrative office eventually by the state secretary. Whereas there is no existence of the superior administrative authoritites in the organizational structure of the state administration, the same body that issued challenged decision decides on it. It is obvious, that appeal procedure contains a lot of variances and peculiarities, which the thesis points out, compared to the appeal procedure. The remostrance is limited to one paragraph and its five sections by the valid and effective legislation of the Administrative Code. The more this relatively brief provision often remains unkonwn to the general public, the more attention it attracts among the experts. The thesis aims to give a comprehensive explanation of the remonstrance and its proceedings and subsequent evaluation of the effectiveness of the applicable legislation and to suggest own creative solution of the examined subject. Further, this thesis aims to answer the...
Territorial self-government in a comparative perspective: Japan and the Czech Republic
Křížek, Jakub ; Kopecký, Martin (advisor) ; Svoboda, Petr (referee)
The purpose of my research is to analyse the territorial self-government in Japan and the Czech Republic from a comparative perspective and provide an outline of the main differences and similarities between the legal conception of local and regional self- government in these two countries. The thesis is composed of ten chapters, each of them dealing with some typical features of the Japanese self-government, such as the constitutional foundations, the personal and territorial dimension of Japanese prefectures and municipalities etc. While the odd chapters are mainly analytical, the even chapters focus on a more detailed comparison of the aforementioned legal issues with their respective equivalents in the Czech law. The first two chapters deal with the question of the constitutional foundations of the territorial self-government in both countries, finding great similarities in both legal conceptions, the main difference being the express provision for direct election of mayors and governors in the Japanese one. Subsequently, the author gives an overview and detailed analysis of the so called basic elements of the self-government, namely population, territory and legal personality including the right to self-government. While the fundamental features of them may overlap in both countries,...
The effect of ethinylestradiol on Na+, K+ - ATPase
Kettnerová, Karolína ; Svoboda, Petr (advisor) ; Driák, Daniel (referee)
This diploma thesis is oriented to analysis of physiological effect of synthetic estrogen ethinylestradiol (EE), which represents the main component of steroid-based substance used in hormonal contraception. From wide range of physiologically important protein molecules, which might be effected by this steroid, thesis focuses to the study of the sodium plus potassium activated, magnesium dependent adenosinetriphosphatase (Na+, K+ - ATPase), which is selectively inhibited by cardiac glycosides such as ouabain (g strophantine). Na+, K+ - ATPase represents an important plasma membrane bound enzyme, which catalyzes the active transport of sodium and potassium across plasma membrane. In the first part of this work, Na+, K+ - ATPase was determined by binding of radioactively labeled selective inhibitor of this enzyme [3H]ouabain, used for this purpose. In the second part of this work, plasma membrane fluidity was analyzed by steady-state fluorescence anisotropy of DPH. The effect of EE on [3H]ouabain binding was studied first under in vitro conditions by using human embryonic kidney cells (HEK293) which were cultivated for 24 hours in the presence of EE in tissue culture medium. Second, the effect of EE was also studied under in vivo conditions, by subcutaneous application of EE to the female rats of Wistar...

Territorial decision-making processes
Jiránek, Vojtěch ; Svoboda, Petr (advisor) ; Vedral, Josef (referee)
This dissertation is concerned with Building Code, No. 183/2006 Sb., and especially in its amendment No. 350/2012 Sb. It specializes in territorial procedure itself which is covered in sections 84 till 96a of Building Code. This dissertation tries to find out what pros and cons of territorial procedure are after the aforementioned amendment. Chapter one lays down the basic terms and documents of territorial procedures on state ("Politika územního rozvoje"), regional ("Zásady územního rozvoje") and urban ("Územní plán") levels. It also covers special institute of "Regulační plan" which can be used on regional and/or urban level. Other parts of chapter one are "Územní rozhodnutí" which specifies the type of given intention in given place and "Veřejnoprávní smlouva" which can be translated as "public (law) contract". This contract enables applicants to make their intention come to effect on their own strength with Building Authority only checking the final state of the contract. At the end this contract has the same power and authority as standard legal act by the Building Authority. Chapter two is the crucial part of this dissertation. It takes separately individual instruments of territorial procedures that are concerned with positioning particular intention(s) and the procedure that is given to this...
Public law restrictions of ownership rights to buildings and the conservation systém
Šimek, Jiří ; Staša, Josef (advisor) ; Svoboda, Petr (referee)
The aim of the thesis is to analyze the public law restrictions to property rights and landmark preservation. At first it introduces the situation of cultural heritage in Czech Republic. Then it focuses on the issue of public interest in landmark preservation. Then is the thesis concerned on special forms of monument preservation. The paper analyzes the duties to take care of landmarks and the restrictions of property rights, especially of the rights to dispose the buildings, to use them, to renew them and to demolish them. Then it surveys how to force the owner to fulfill his duties, when he fails to comply with the restrictions and duties. The last topic is about compensation for the public law restrictions and about the case law, which helps us to distinguish between inner and outer restriction of property rights. At the end, it sums up the main problems of today landmark preservation system, especially the lack of efficient enforcement tools against the owner who doesn't fulfill his duties.
