Host factors involved in Rous Sarcoma Virus replication
Štafl, Kryštof ; Svoboda, Jan (advisor) ; Horníková, Lenka (referee)
Rous sarcoma virus (RSV) takes a place of honor among retroviruses. Research of RSV allows us to uncover the secret of the origin and evolution of life, the mechanisms of tumorigenesis and the interaction between viruses and their hosts. Viruses are not able to replicate themselves without host cells. They exploit a number of cellular pathways and factors and they can reprogram the cells to produce great amounts of viral progeny. They exert pressure on their host that leads to developement of new types of cellular proteins, which then results in resistance of the cells. This thesis focuses on the host factors involved in the RSV replication cycle. It summarizes the current knowledge about the replication cycle of retroviruses and the host factors that are necessary for productive infection: cellular receptors, endocytic and secretory pathways, nuclear transport, proteosynthesis, replication of proviruses and its stimulation. The restriction mechanisms of cells are also taken into account. The current knowledge about RSV is compared with facts on mammalian retroviruses and gaps in research are highlighted. The influence of the host cell factor absence, host specificity and cellular permissiveness are correlated and discussed.

This bachelors thesis deals with the synthesis of bis(terpyridyl)oligofluorenes, that are suitable for the preparation of supramolecular polymers. The main aim was synthesis and characterisation of 7,7'-bis(terpyrid-4'-yl)-9,9,9',9'-tetrakis(6'-bromohexyl)-2,2'-bifluorene. As a starting substance was used commercially available 2-bromofluorene made by Sigma- Aldrich. In the preparation of the final ligand was used three reaction schemes, during the several steps used in the Suzuki coupling, or variations thereof.
Procyclicality of Bank Lending and Provisioning Behavior
Svoboda, Jan ; Šopov, Boril (advisor) ; Lešanovská, Jitka (referee)
The aim of this paper is to investigate the procyclical behavior of banks in terms of lending and loan loss provisioning, and its dynamics with regard to the adoption of the Basel II capital regulation. Using bank-level and country-level panel data spanning from 1996 to 2013 we answer this question for the OECD and BRIC countries. We find a positive effect of bank capitalization on loans growth, which, perhaps due to the recent financial crisis, weakened after 2008. Together with evidence of income smoothing and capital management we also find strong cyclical behavior of banks in terms of loan loss provisioning. At the same time, we do not find any robust changes to this behavior after the introduction of the Basel II capital regulation. We fill a gap in the empirical literature as there has been hardly any research done on changes brought forward by the adoption of the Basel II capital regulation. The results may be therefore of interest for regulators and other professionals. Moreover, we use in our analysis data for BRIC countries, which have been often neglected.
Preparation and properties of building blocks of specialty polymers
Šichová, Kristýna ; Svoboda, Jan (advisor) ; Sedláček, Jan (referee)
This Diploma Thesis presents results obtained by solution of two partial projects: a) Preparation of monomers from renewable sources using metathesis and tandem hydrogenation catalyzed with ruthenium compounds - project solved during my Erasmus stay at the Université de Rennes 1 in France; b) Preparation and properties of ,-bis(tpy)quarterthiophene oligomers carrying ionic side groups as oligomonomers for polyelectrolyte conjugated dynamers - project solved at the Department of Physical and Macromolecular Chemistry, Faculty of Science, Charles University in Prague. Project a): Self-metathesis of 1,2-epoxyhex-5-ene (but-3-enyloxirane) and its cross-metathesis with methyl acrylate and acrylonitrile catalyzed with ruthenium compounds as well as tandem design of these metatheses and consecutive hydrogenation of their products by gaseous hydrogen have been optimized. The following influences have been studied and tuned: (i) type of the catalyst (Grubbs, Hoveyda, Zhan) and its concentration and method of dosing, (ii) concentration of reactants and additives, (iii) type of solvent, and (iv) reaction temperature. Reactions were monitored by the GC, GC and MS methods and the products were characterized by the NMR method. Methyl 6,7-epoxyheptanoate (methyl 5-oxiranylpentanoate) obtained by the tandem...
Direct arylation and its application in supramolecular polymer synthesis
Loukotová, Lenka ; Svoboda, Jan (advisor) ; Drahoňovský, Dušan (referee)
This thesis is divided into two parts. In the first part there is reported palladium- catalysed regioselective direct arylation of benzofurans with arylsulfonyl chlorides as the coupling partner to C2 carbon. The best reaction conditions of this reaction were investigated, then the influence of various arylsulfonyl chloride substituents to the reaction and finally it is reported direct arylation of C2-arylated benzofuran to C3 carbon to give diarylated benzofuran with different aryl substituents. In the second part, there is reported the synthesis of 2-(hydroxymethyl)-5,7-bis{5- [(2,2':6',2''-terpyridine)-4'-yl]thiophen-2-yl}-2,3-dihydrothieno[3,4-b][1,4]dioxin and modification of its sidechain. As a starting material of the synthesis it was used commercially available 4'-bromo-(2,2':6',2''-terpyridine). The synthetic sub-steps included Suzuki cross- coupling, bromination and C-H bond activation. Then it was also reported the complexation of synthesized oligomer with zinc and iron cations.
Flexibility in spatial cognition of rat
Staňková, Anna ; Svoboda, Jan (advisor) ; Telenský, Petr (referee)
5 Abstract Cognitive flexibility is an ability to adapt a behavior according to a dynamic changes in an environment. It belongs to executive functions, along with the processes of working memory, attention, motivation and others. Flexibility is important in the process of changing "stimulus-response" contingency at the time when the initial stimulus no longer leads to reward. According to the nature of the changes we can distinguish three types of cognitive flexibility: reversal learning, intradimensional set shifting and extradimensional set shifting. The processes of cognitive flexibility take place in fronto-striato-thalamic circuit with numerous connections to other brain regions, especially the limbic system. Deficit in cognitive flexibility belongs among cognitive symptoms of schizophrenia, a psychiatric illness we studied in the experimental part of this work using its analogue in rat. The aim of the experimental work was to test cognitive flexibility deficit in the rat model of schizophrenia-like behavior in the Carousel maze in four versions of task with different cognitive load. Animal model of schizophrenia was induced by acute administration of non-competitive NMDA receptor antagonist MK-801 at doses of 0.05 and 0.10 mg/kg i.p. Reversal learning was not damaged in this type of task following the...
The nedia image of the Czech republic in the english-speaking world
Svoboda, Jan ; Trampota, Tomáš (advisor) ; Dvořák, Tomáš (referee)
The title of the presented thesis is The Media Image of the Czech Republic in the English-speaking World. The media image of the Czech Republic is analyzed in three different English-written newspapers. The chosen newspapers are USA Today, The Daily Mail and The Australian. The overall goal of the thesis is to compare media images which are constructed by defined periodicals and it is reached by checking of the hypotheses which are defined in the introduction and thanks to the comparison of data gathered during the research. The theoretical part of the thesis anchors the work itself within contemporary theories and academic approaches. The news production with international news flows included are described in the theoretical chapter along with stereotypes and their relation to media images. The whole chapter is ended by subchapter about hard and soft news and subchapter about evaluation in media. The analytical part of the thesis begins with a description of a used research method and a definition of used data. Subsequently, there is a detailed description of used variables. The very last part of the whole paper presents complex results of the research, the hypotheses are proved or falsified and the differences among the media image of the analyzed newspapers are described.
The midlife crisis in the marriage of Christians
Svoboda, Jan ; Bartůšková, Ludmila (advisor) ; Opatrný, Aleš (referee)
The bachelor's thesis "The midlife crisis in the marriage of Christians" first describes in what lies the mid-life crisis in human life. It takes notice of general causes and differences in experiencing the crisis between men and women. This period is demanding for the relations between the spouses. Couples who live the Christian faith are not immune against crises, however it might be a challenge for them. They also must seek the ways how to come out of the crisis together. They have to fight for their faith and their marriage, which is in the stage of requiring a new qualitative deepening of the relationship. Wider possibilities of assistance are available to the Christian spouses, thanks to their faith. This thesis combines a psychological view of the crisis in the mid-life with the spiritual perspective of Christian spouses who are enduring this crisis, but who can draw strength for such a period from their faith.
Testing of spatial orientation in leopard gecko ( Eublepharis macularius)
Voňavková, Monika ; Landová, Eva (advisor) ; Svoboda, Jan (referee)
The ability of the spatial orientation of the reptiles is not fully researched yet. It is unknown if reptiles use only simple types of navigation, for example cue learning (one key mark), or more difficult types, for example allothetic navigation (combination of marks) or cognitive map (mental representation of the area). The main aim of this thesis was examine the possibility of the spatial orientation in lizard leopard gecko (Eublepharis macularius) in modificated Morris water maze (MWM). Design of the experiments was based on Parallel Map Theory, a theory which describes using spatial information from more navigation frames. The leopard gecko preferences of orientation frames were tested . That means the ability to combinate the spatial information from the bearing maps (one mark and gradient) and the sketch maps (local marks). This ability is prerequisite for creating the integrated map. The research was also focused on the significance of the simple cue learning strategy and cognitive more difficult allothetic navigation for the orientation of leopard gecko. Leopard geckos can use the combination of information from the navigation frames, which indicate the possibility of integrated map creation. Usage of navigation strategies is individual. Keywords: reptiles, leopard gecko, spatial...
